Metabolic profile of fish muscle tissue changes with sampling method, storage strategy and time.

Unstable tissue components (metabolites) are not easily captured and evaluated by traditional metabolomics methods. In this study, a comprehensive investigation of various sampling methods and storage conditions on the metabolomic profile of fish muscle was performed based on in vivo and ex vivo sampling. The GlobalStd algorithm and structure/reaction directed analysis under a linear mixed model were used to investigate the distinctive influences of these factors on the metabolomic profiles of fish tissue obtained via untargeted LC-MS analysis. Immediate analysis of samples yielded different metabolomic profiles compared to that of stored samples. Storage time was found to affect the metabolomic profile in a complex way, whereas storage temperature was shown to not substantially change this pattern. At the reaction level, metabolites involved in homologous series with butylation were shown stable during storage. Overall, our findings demonstrate that immediate instrumental analysis after in vivo solid phase microextraction (SPME) sampling and a reverse time series experimental design should be the preferred approaches for metabolomic profiling if unstable compounds are of interest.

In this study, we attempt to illustrate fossil vertebrate dental tissue geochemistry and, by inference, its extent of diagenetic alteration, using quantitative, semi-quantitative and optical tools to evaluate bioapatite preservation. We present visual comparisons of elemental compositions in fish and plesiosaur dental remains ranging in age from Silurian to Cretaceous, based on a combination of micro-scale optical cathodoluminescence (CL) observations (optical images and scanning electron microscope) with minor, trace and rare earth element (REE) compositions (EDS, maps and REE profiles), as a tool for assessing diagenetic processes and biomineral preservation during fossilization of vertebrate dental apatite. Tissue-selective REE values have been obtained using laser ablation-inductively coupled plasma-mass spectrometry (LA-ICP-MS), indicating areas of potential REE enrichment, combined with cathodoluminescence (CL) analysis.

In viviparous black rockfish (Sebastes schlegelii), the kidney of reproductive-phase males actively produces lipocalin-type prostaglandin D synthase homolog (LPGDSh) protein, which is presumably involved in intersexual communication when emitted in the urine. The present study was undertaken to discover whether androgens and their nuclear receptors (Ars) are engaged in regulation of renal LPGDSh protein synthesis in black rockfish. Quantitative real-time polymerase chain reaction, in conjunction with immunohistochemistry and highly sensitive enzyme-linked immunosorbent assay, revealed that intra-abdominal administration of a synthetic androgen, 17α-methyltestosterone (MT), to juvenile black rockfish induced their renal expression of LPGDSh transcript and protein.

Antarctic krill (Euphausia superba), which is rich in astaxanthin, has been widely utilized as a dietary supplement in fish aquaculture. Our study was to feed juvenile leopard coral grouper (Plectropomus leopardus) a diet containing 50 % Antarctic krill, revealing significant body color differentiation between a reddened group (BKR) and a non-reddened group (BKB), followed by comparative analysis with the control group (BCon) without krill supplementation. Histological analysis and carotenoid content in the liver and intestine were differentially regulated in color-differentiated individuals.

Adenoid cystic carcinomas (AdCC) of salivary gland origin have long been categorized as fusion-defined carcinomas owing to the almost universal presence of the gene fusion MYB::NFIB, or less commonly MYBL1::NFIB. Coastal fish populations are threatened by multiple anthropogenic impacts, including the accumulation of industrial contaminants and the increasing frequency of hypoxia. Some populations of the Atlantic killifish (Fundulus heteroclitus), like those in New Bedford Harbor (NBH), Massachusetts, USA, have evolved a resistance to dioxin-like polychlorinated biphenyls (PCBs) that may influence their ability to cope with secondary stressors.
