Histamine H receptor gene variants associated with drug abuse in patients with cocaine use disorder.

Published: November 2020

Background: Preclinical work revealed significant interactions between ligands of the histamine H receptor and different drugs of abuse. In the case of psychostimulants, the results reported are somewhat controversial and human data are still scarce, despite the fact that an inverse agonist of the H receptor (pitolisant) has reached the market after approval for the treatment of narcolepsy.

Aims: We have studied associations between histamine H receptor gene variants and cocaine use disorder to increase the knowledge of the possible involvement of histamine H receptor in drug abuse.

Methods: Seven single nucleotide polymorphisms of the histamine H3 receptor gene were genotyped by using a multiplexing assay in 248 samples of subjects with cocaine use disorder and 500 randomized samples of subjects representative of the Spanish population.

Results: The study of the epidemiological information associated to the samples revealed that subjects with cocaine use disorder broadly abused alcohol, tobacco and cannabinoids. Two single nucleotide polymorphisms (rs3787430 and rs74627870) were found significantly associated with the occurrence of addiction and one more (rs13042865) was specifically related to the severity of cocaine dependence within drug abusers.

Conclusions: The associations found in this study further extend the hypothesis that histamine H receptor function could be relevant in drug abuse in general and cocaine addiction in particular.

The role of rodent behavioral models of schizophrenia in the ongoing search for novel antipsychotics.

Expert Opin Drug Discov

January 2025

Centro de Investigación en Reproducción Animal, Universidad Autónoma de Tlaxcala - CINVESTAV Tlaxcala, Tlaxcala, México.

Introduction: Existing pharmacotherapies for schizophrenia have not progressed beyond targeting dopamine and serotonin neurotransmission. Rodent models of schizophrenia are a necessary tool for elucidating neuropathological processes and testing potential pharmacotherapies, but positive preclinical results in rodent models often do not translate to positive results in the clinic.

Areas Covered: The authors reviewed PubMed for studies that applied rodent behavioral models of schizophrenia to assess the antipsychotic potential of several novel pharmacotherapies currently under investigation.

Integrating Machine Learning and Pharmacophore Features for Enhanced Prediction of H1 Receptor Blockers.

Introduction: Histamine Type I Receptor Antagonists (H1 blockers) are widely used to mitigate histamine-induced inflammation, particularly in allergic reactions. Histamine, a biogenic amine found in endothelial cells, vascular smooth muscle, bronchial smooth muscle, and the hypothalamus, is a key player in these responses. H1 blockers are essential in cough syrups and flu medications and are divided into two generations: first-generation H1 blockers, which are sedating and have numerous side effects, and second-generation blockers, which are non-sedating and generally less toxic but may still exhibit cross-reactivity with other receptors.

Proton pump inhibitors and all-cause mortality risk among cancer patients.

World J Clin Oncol

January 2025

Department of Supportive Oncology, Atrium Health Levine Cancer, Charlotte, NC 28204, United States.

Background: Proton pump inhibitors (PPIs) are widely used, including among cancer patients, to manage gastroesophageal reflux and other gastric acid-related disorders. Recent evidence suggests associations between long-term PPI use and higher risks for various adverse health outcomes, including greater mortality.

Aim: To investigate the association between PPI use and all-cause mortality among cancer patients by a comprehensive analysis after adjustment for various confounders and a robust methodological approach to minimize bias.

Lenacapavir is a potent, long-acting HIV-1 capsid inhibitor used in combination with other antiretrovirals to treat HIV-1 infection. The pharmacokinetics of orally administered drugs may be affected by food intake or coadministration of acid-reducing agents (ARA). Two Phase 1 studies were conducted on healthy participants to evaluate the effect of food and the impact of the histamine H-receptor antagonist famotidine in parallel cohorts.
