Cadmium (Cd) is an environmental toxicant and a human carcinogen. Several studies show an association of Cd exposure to the development of breast cancer. Previously, we have transformed the immortalized non-tumorigenic cell line MCF-10A with Cd and have demonstrated that the transformed cells have anchorage independent growth. In a separate study, we showed that transformation of the immortalized urothelial cells with the environmental carcinogen arsenite (As) results in an increase in expression of genes associated with the basal subtype of bladder cancer. In this study, we determined if transformation of the MCF-10A cells with Cd would have a similar effect on the expression of basal genes. The results of our study indicate that there is a decrease in expression of genes associated with keratinization and cornification and this gene signature includes the genes associated with the basal subtype of breast cancer. An analysis of human breast cancer databases indicates an increased expression of this gene signature is associated with a positive correlation to patient survival whereas a reduced expression/absence of this gene signature is associated with poor patient survival. Thus, our study suggests that transformation of the MCF-10A cells with Cd produces a decreased basal gene expression profile that correlates to patient outcome.

Understanding the molecular energy metabolism of single cells in the nucleolus stress response induced by mild-photothermal therapy (mPTT) is of great importance for investigating the photothermal lethal mechanism. Herein, we successfully fabricated a "turn-on"-type fluorescent nanoprobe based on the fluorescently labeled aptamers (FAM-ATP-apt and Cy3-GTP-apt) and TiCT MXene. When the adapters on the nanoprobes bonded to intracellular ATP and GTP, the fluorescence of the nanoprobes was restored.

2-arachnadoyl glycerol (2-AG) is one of the most common endocannabinoid molecules with anti-proliferative, cytotoxic, and pro-proliferative effects on different types of tumors. Typically, it induces cell death via cannabinoid receptor 1/2 (CB1/CB2)-linked ceramide production. In breast cancer, ceramide is counterbalanced by the sphingosine-1-phosphate, and thus the mechanisms of 2-AG influence on proliferation are poorly understood.

Accompanying the occurrence of inflammatory reaction to release cytokines, pyroptosis can activate an immune response for resistance against cancer. Consequently, elevated levels of cytokines released by cancer cells are highly correlated with the effectiveness of cancer treatment. Herein, a noninvasive surface-enhanced Raman spectroscopy (SERS) immunosensor was developed to sensitively and specifically measure the tumor necrosis factor-α (TNF-α), a proinflammatory cytokine, during the cell pyroptosis process.

Recent advances in cancer therapy have been made possible by monoclonal antibodies, domain antibodies, antibody drug conjugates, The most impact has come from controlling cell cycle checkpoints through checkpoint inhibitors. This manuscript explores the potential of a series of novel -benzyl isatin based hydrazones (5-25), which were synthesized and evaluated as anti-breast cancer agents. The synthesized hydrazones of -benzyl isatin were screened against two cell lines, the MDA-MB-231 breast cancer cell line and the MCF-10A breast epithelial cell line.

In this study, the copper(II) complex [Cu(chromoneTSC)Cl]•0.5HO•0.0625CHOH (where chromoneTSC = -Ethyl-2-((4-oxo-4H-chromen-3-yl)methylene)-hydrazinecarbothioamide) was synthesized and characterized; then used to carry out studies in combination with berberine chloride (BBC).
