Rheumatoid arthritis (RA) is a systemic autoimmune disorder with both articular and extra-articular manifestations, including rare pulmonary complications. We report a case of a 65-year-old male with long-standing RA who developed multiple cavitary pulmonary nodules following prolonged leflunomide therapy. Diagnostic evaluation excluded infectious, neoplastic, and autoimmune causes.

In silico trials for drug safety assessment require many high-fidelity 3D cardiac simulations to predict drug-induced QT interval prolongation, which is often computationally prohibitive. To streamline this process, we developed sex-specific emulators for a fast prediction of QT interval, trained on a dataset of 900 simulations. Our results show significant differences between 3D and 0D single-cell models as risk levels increase, underscoring the ability of 3D modeling to capture more complex cardiac responses.

Minocycline-induced lupus.

BMJ Case Rep

December 2024

Division of Infectious Diseases, Department of Internal Medicine, University of Iowa, Iowa City, Iowa, USA.

Antibiotics are indicated for inappropriately prolonged durations not only for infectious diseases but also for non-infectious dermatologic conditions such as acne vulgaris, but inappropriate use is not uncommon. Inappropriate antibiotic prescription can contribute to the emergence of multidrug-resistant organisms, patient harm such as adverse drug events and increased healthcare costs. We experienced a case involving a late adolescent with acne vulgaris who had been treated with minocycline for more than 2 years, leading to the development of drug-induced lupus, attributed to minocycline.

(1) Background: Drug-induced liver injury is a prevalent global health concern that necessitates urgent development of safe and effective treatment options for patients. Drug-carrying nanoparticles have garnered significant attention for dis-ease treatments due to their capacity to enhance drug solubility, provide drug protection, and prolong release duration, thereby improving drug bioavailability and increasing therapeutic efficacy. We initially present a nanostructured carrier incorporating glycyrrhetinic acid and transferrin.

The assessment and aetiology of drug-induced ischaemic priapism.

Int J Impot Res

December 2024

Associate Professor in the Division of Urology, Department of Surgery, School of Clinical Medicine, Faculty of Health Sciences, University of the Witwatersrand, Johannesburg, South Africa.

Ischaemic priapism is a urological emergency characterised by a prolonged, painful erection unrelated to sexual stimulation. While several aetiological factors contribute to this condition, the pharmacological causes have gained significant attention in recent years. This narrative review aims to comprehensively assess ischaemic priapism, specifically focusing on its pharmacological aetiology.
