Ultrashort cationic lipopeptides (USCLs) are considered to be a promising class of antimicrobials with high activity against a broad-spectrum of microorganisms. However, the majority of these compounds are characterized by significant toxicity toward human cells, which hinders their potential application. To overcome those limitations, several approaches have been advanced. One of these is disulfide cyclization that has been shown to improve drug-like characteristics of peptides. In this article the effect of disulfide cyclization of the polar head of -palmitoylated USCLs on in vitro biological activity has been studied. Lipopeptides used in this study consisted of three or four basic amino acids (lysine and arginine) and cystine in a cyclic peptide. In general, disulfide cyclization of the lipopeptides resulted in peptides with reduced cytotoxicity. Disulfide-cyclized USCLs exhibited improved selectivity between sp., Gram-positive strains and normal cells in contrast to their linear counterparts. Interactions between selected USCLs and membranes were studied by molecular dynamics simulations using a coarse-grained force field. Moreover, membrane permeabilization properties and kinetics were examined. Fluorescence and transmission electron microscopy revealed damage to cell membrane and organelles. Concluding, USCLs are strong membrane disruptors and disulfide cyclization of polar head can have a beneficial effect on its in vitro selectivity between sp. and normal human cells.

A novel and efficient strategy for the direct synthesis of 3-arylthioquinoline derivatives via radical induced tandem cyclization of propargylamines with diaryl disulfides was developed. This protocol undergoes a cascade sulfuration/ cyclization/ oxidation/ aromatization pathway to afford the desired products in a broad substrate scope using readily available starting materials under mild conditions. Based on this strategy, we further modified 3-arylquinolines to obtain two novel deep blue fluorescent molecules, QLSCz and QLSTCz, with good optical properties through two-step synthesis by oxidation and electron donor modification.

The photoacid-catalyzed synthesis of 2-deoxy glycosides is presented using stable glycosyl -[1-(-MeO-Phenyl)vinyl]benzoate (PMPVB) donors and employing the eosin Y and diphenyl disulfide (PhSSPh) catalytic system in the presence of blue LED lights. The remote activation of the alkene functionality under the photoacid catalysis followed by a 5-- cyclization led to the generation of oxocarbenium ions that were trapped to provide the glycosylated products in excellent yields and decent selectivities under mild conditions. This method is also useful for the photoacid-catalyzed synthesis of -methoxybenzyl-alkyl ethers.

We have de novo designed a helix-loop-helix (HLH) peptide (MW: ∼4,500), constructed phage-displayed libraries, and screened the libraries against a variety of disease-related proteins to successfully obtain molecular-targeting HLH peptides. The next essential step in developing HLH peptides into therapeutics involves affinity engineering to optimize binding affinity and specificity.

In this report, we have established a novel and efficient method for selectively synthesizing either vinyl sulfides or 2-methylbenzothiazoles from the reaction of CaC and disulfides. The selective synthesis of these two distinct products can be controlled by simply adjusting the amount of KS. The underlying reaction mechanism has been thoroughly investigated through control experiments, HRMS, and FTIR, which collectively support the pivotal role of a trisulfur radical anion.
