Conjugation, that is, covalent linkage, to immunological proteins is a common strategy to address the low immunogenicity issue of carbohydrate antigens in vaccine development. This chapter describes an easy and efficient method for oligosaccharide-protein conjugation employing dicarboxylic acid linkers. In this regard, a free amino group is introduced to an oligosaccharide antigen to facilitate coupling with the bifunctional linker upon reaction with its corresponding disuccinimidyl ester. The resultant monosuccinimidyl ester of the oligosaccharide antigen then reacts with the free amino groups of a carrier protein to provide the desired oligosaccharide-protein conjugate.

Rotaviruses, non-enveloped viruses with a double-stranded RNA genome, are the leading etiological pathogen of acute gastroenteritis in young children and animals. The P[11] genotype of rotaviruses exhibits a tropism for neonates. In the present study, a binding assay using synthetic oligosaccharides demonstrated that the VP8* protein of P[11] porcine rotavirus (PRV) strain 4555 binds to lacto-N-neotetraose (LNnT) with the sequence Galβ1,4-GlcNAcβ1,3-Galβ1,4-Glc, one of the core parts of histo-blood group antigen (HBGA) and milk glycans.

Globo H, a specific carbohydrate antigen overexpressed on various human malignancies, has attracted considerable interest as an antigenic target for anticancer vaccine development. Despite several Globo H-based carbohydrate vaccines that have been designed, efficient access to Globo H hexasaccharide antigen and development of powerful adjuvants for enhancing antitumor immunity remain challenging. Herein, we reported a streamlined chemoenzymatic approach to prepare this hexasaccharide antigen, relying on chemical synthesis of Gb5 pentasaccharide by a stereoconvergent [2+3] strategy and subsequent enzymatic α-fucosylation to easily install α1,2-fucose residue.
