Isothermal Titration Calorimetry (ITC) elucidates the thermodynamic profile (ΔH, ΔS, ΔG, Ka, and stoichiometry) of binding and dissociation reactions in solution. While ITC has primarily been used to investigate the thermodynamics of interactions between biological macromolecules and small molecules, it has become increasingly common for measuring binding interactions between synthetic polymers and small molecules, ions, or nanoparticles. This tutorial review describes applications of ITC in studying synthetic macromolecules and provides experimental guidelines for performing ITC experiments. We also highlight specific examples of using ITC to study soft matter, then discuss the limitations and the future of ITC in this field.

We report the synthesis of a series of detergents with a lactobionamide polar head group and a tail containing four to seven perfluorinated carbon atoms. Critical micellar concentrations (CMCs) were determined using isothermal titration calorimetry (ITC) and surface tension (SFT) measurements, showing a progressive decrease from 27 mM to about 0.2 mM across the series.

Hepatic ischemia-reperfusion injury (IRI) poses a significant threat to clinical outcomes and graft survival during hemorrhagic shock, hepatic resection, and liver transplantation. In this study, a supramolecular fluorescent material was constructed by using double-cavity twisted cucurbit[14]uril (tQ[14]) and positively charged Astrazon Pink FG (APFG) based on the non-covalent host-guest interaction for the first time. The thermodynamic parameters of the APFG@tQ[14] in aqueous solution were determined by isothermal titration calorimetry (ITC), the results indicated that the spontaneous assembly of APFG@tQ[14] is mainly driven by enthalpy. The intramolecular charge transfer (ICT) effect induced the APFG@tQ[14] probe to emit a strong orange-red fluorescence.
