, the causal agent of apple Valsa canker, produces cankers, resulting in the death of infected tissues and eventually the entire tree. Because of the long latent period of the disease, it is necessary to develop a rapid, sensitive, and reliable field-based assay to effectively diagnose apple Valsa canker when the plant is still symptomless. Loop-mediated isothermal amplification (LAMP) is a novel detection method that synthesizes a large amount of DNA and produces the visible byproduct (magnesium pyrophosphate) without conventional thermal cycling. Six LAMP primers were designed to target a species-specific region of the elongation factor-1α sequence, which can be completed at 61°C in 60 min. A positive result is indicated by color change after the intercalating dye SYBR Green I is added. The specificity of the LAMP was validated with DNA from 45 representative isolates of and closely related species , , and . The sensitivity of the LAMP was determined to be 1 ng of DNA or as few as 10 spores. Because the assay does not require expensive equipment or specialized techniques, the LAMP-based diagnostic method can be applied under field conditions to more precisely and efficiently access disease pressure in apple orchards.

Valsa canker, caused by fungal pathogens in Valsa species, is a fungal disease of apple and pear growing in China and even in Asia. Malectin-like kinases play crucial roles in plant recognition of the pathogen-induced signals and subsequent activation of partially host immune responses. However, the role of MEDOS1 (MDS1), a Malectin-like kinase, in plant immunity has not yet been extensively explored.

In search of novel natural product-based fungicides,twenty-four cuminic acid derivatives containing thiourea group were synthesized and characterized by 1H-NMR, 13C-NMR and HRMS spectra. The anti-fungi/oomycete activity bioassay showed that the some of the title compounds displayed moderate to good anti-fungi activity against Botryosphaeria dothidea (B. dothidea), Sclerotinia sclerotiorum (S.

To mitigate the impact of traditional chemical pesticides on environment, and achieve sustainable crop protection, 24 eco-friendly rosin-based sulfonamide derivatives were synthesized and developed. The in vitro activity assessment showed that compound 4X (Co. 4X) exhibited excellent fungicidal activity against V.

Apple Valsa canker (AVC) caused by the Ascomycete Valsa mali, seriously constrains the production and quality of apple fruits. The symptomless incubation characteristics of Valsa mali make it highly challenging to detect AVC at an early infection stage. After infecting the wound of apple bark, the pathogenic hyphae of AVC will expand and colonize the phloem tissue.
