We have developed a combination of light scattering techniques to study and characterize droplets of an ultrasonic spray coater in flight. For this economically relevant spray coater, there is so far no reliable technique to systematically adjust the experimental parameters. We have combined photon correlation spectroscopy and turbidimetry to determine the size and speed of the droplets depending on parameters of the printing process as shroud gas pressure, flow rate, and atomizing power. Our method will allow us to predetermine these parameters to control the properties of the coated films as, e.g., thickness from tens of nanometers to micrometers.

In situ calcium pectinate-coated pellets were proposed by applying an alternate coating method to drug-layered pellets to achieve colon-specific drug delivery. Solution layering of metroprolol tartrate, a water-soluble model drug, on inert core pellets was achieved using a centrifugal granulator followed by successive alternate coating with pectin and calcium chloride layers using a fluidized bed bottom spray coater. Oral solid dosage forms that contain APIs in the amorphous state have become commonplace because of many drug substances exhibiting poor water solubility, which negatively impacts their absorption in the human GI tract. While micronization, solvent spray-drying, and hot-melt extrusion can address solubility issues, spray coating of the APIs onto beads and tablets offers another option for producing amorphous drug products. High-level comparisons between bead and tablet coating technologies have the potential for simpler equipment and operation that can reduce the cost of development and manufacturing.
