Published: December 2020

The transition-metal-free cross-coupling of alkyl or aryl electrophiles by using tertiary benzylic organoboronates is reported. This reaction involves the generation of tertiary alkyl anions from organoboronates in the presence of an alkoxide base and then their substitution reactions. This protocol allows the simple and efficient construction of quaternary carbon centers.

-Ge/B 1,1-Hydroboration of Alkynylgermanes with 9-BBN.

A 1,1-hydroboration of alkynylgermanes with unique -Ge/B stereochemistry under transition-metal-free conditions is reported. Mechanistic studies suggest that a pathway involving α-boration followed by a stepwise 1,2-Ge/H shift on the intermediate structurally lies between an alkyne-Ge π complex and a typical vinyl cation. The resulting Ge/B bimetallic modules, along with a /Ge/B trimetallic variant, can be conveniently transformed into trisubstituted olefins through iterative divergent cross-coupling.

Functionalization of Pyridines at the C4 Position via Metalation and Capture.

The functionalization of pyridines at positions remote to the N-atom remains an outstanding problem in organic synthesis. The inherent challenges associated with overriding the influence of the embedded N-atom within pyridines was overcome using n-butylsodium, which provided an avenue to deprotonate and functionalize the C4-position over traditionally observed addition products that are formed with organolithium bases. In this work, we show that freshly generated 4-sodiopyrdines could undergo transition metal free alkylation reactions directly with a variety of primary alkyl halides bearing diverse functional groups.

Reductive deaminative cross-coupling of alkyl bistriflimides enabled by electrocatalysis.

We present a versatile nickel-electrocatalytic deaminative cross-coupling platform for the efficient construction of C(sp)-C(sp) and C(sp)-C(sp) bonds from readily available alkyl bistriflimides. This methodology involves the assembly of two leaving groups on alkyl amines to form alkyl bistriflimides, followed by their effective coupling with a wide range of alkyl halides, alkyl pseudohalides, aryl halides, and alkenyl halides under electrochemical reductive conditions. Moreover, the successful application of electrochemical reductive relay cross-coupling and transition metal-free cross-electrophile coupling further demonstrates the versatility of alkyl bistriflimides as valuable building blocks in organic synthesis.

Coupling of unactivated alkyl electrophiles using frustrated ion pairs.

Laboratorium für Organische Chemie, ETH Zürich, Zürich, Switzerland.

Cross-electrophile coupling reactions have evolved into a major strategy for rapidly assembling important organic molecules. Two readily accessible electrophiles are coupled to form new C-C bonds, providing a key advantage over traditional cross-coupling strategies that require the preformation of reactive organometallic species. Yet, the formation of C(sp)-C(sp) bonds that form the core of nearly all organic compounds remains highly challenging with current approaches, calling for the design of innovative new strategies.

Unveiling the Significance of tert-Butoxides in Transition Metal-Free Cross-Coupling Reactions.

The astounding reactivity of tert-butoxides in transition metal-free coupling reactions is driving the scientific community towards a new era of environmental friendly, as well as cost-effective, transformation strategies. Transition metal-catalyzed coupling reactions generate hazardous wastes and require harsh reaction conditions, mostly at elevated temperature, which increases not only costs but also environmental concerns regarding the methodology. Tert-butoxide-catalyzed/mediated coupling reactions have several advantages and potential applications.
