α-Glucans that were enzymatically synthesized from sucrose using glucansucrase cloned from NRRL B-1118 were found to have a glass transition temperature of approximately 80 °C. Using high-pressure homogenization (~70 MPa), the α-glucans were converted into nanoparticles of ~120 nm in diameter with a surface potential of ~-3 mV. Fluorescence measurements using 1,6-diphenyl-1,3,5-hexatriene (DPH) indicate that the α-glucan nanoparticles have a hydrophobic core that remains intact from 10 to 85 °C. α-Glucan nanoparticles were found to be stable for over 220 days and able to form at three pH levels. Accelerated exposure measurements demonstrated that the α-glucan nanoparticles can endure exposure to elevated temperatures up to 60 °C for 6 h intervals.

A novel aggregation-induced emission (AIE)-based artificial light-harvesting system (LHS) is successfully assembled via the host-guest interaction of bis-naphthylacrylonitrile derivative (BND), water-soluble pillar[5]arene (WP5), and sulforhodamine 101 (SR101). After host-guest assembly, the formed WP5⊃BND complexes spontaneously self-aggregated into WP5⊃BND nanoparticles (donors) and SR101 (acceptors) is introduced into WP5⊃BND to fabricate WP5⊃BND-SR101 LHS. Introduction: Although photodynamic therapy (PDT) shows considerable potential for cancer treatment due to its precise spatial control and reduced toxicity, effectively eliminating residual cells under hypoxic conditions remains challenging because of the resistance conferred by these cells.
Methods: Herein, we synthesize an amphiphilic PEGylated polyphosphoester and present a nanocarrier (NP) specifically designed for the codelivery of hydrophobic photosensitizer (chlorin e6, Ce6) and hypoxia-activated prodrugs (tirapazamine, TPZ). We investigate the antitumor effect of NP on both cellular and animal level.
