We report a DNA-compatible protocol for synthesizing amides from DNA-bound aldehydes and non-nucleophilic arylamines including aza-substituted anilines, 2-aminobenzimidazoles, and 3-aminopyrazoles. The reactions were carried out at room temperature and provided reasonable conversions and wide functional group compatibility. The reactions were also successful when employing aryl and aliphatic aldehydes. In addition, qPCR and NGS data suggested no negative impact on DNA integrity after the copper-mediated oxidative amidation reaction.

A catalyst-free photochemical N-acylation of azoles with aldehydes has been developed using inexpensive BrCCl3 as the oxidant. This transition-metal- and photocatalyst-free amidation proceeded efficiently with a wide variety of substrates to give the corresponding N-acylazoles, including for the late modification of pharmaceutically active molecules, and on a gram-scale.

Background/aims: Gestational Diabetes Mellitus (GDM) is a common complication during pregnancy, defined as diabetes diagnosed in the second or third trimester, often asymptomatic. This study investigates the therapeutic potential of olive leaf extracts and stem cells in mitigating GDM-induced complications, particularly focusing on renal function, oxidative stress, and pancreatic cell regeneration.
Methods: Measurements were made in gravid female rats with or without intraperitoneal administration of Streptozotocin (35 mg/kg body weight).

The human voltage-gated proton channel (H1) provides an efficient proton extrusion pathway from the cytoplasm contributing to the intracellular pH regulation and the oxidative burst. Although its pharmacological inhibition was previously shown to induce cell death in various cell types, no such effects have been examined in polarized macrophages albeit H1 was suggested to play important roles in these cells. This study highlights that 5-chloro-2-guanidinobenzimidazole (ClGBI), the most widely applied H1 inhibitor, reduces the viability of human THP-1-derived polarized macrophages at biologically relevant doses with M1 macrophages being the most, and M2 cells the least sensitive to this compound.

This study aimed to investigate the protective effect of a novel capsaicinoid glucoside (CG) against HO-induced oxidative stress in HepG2 cells and elucidate its underlying molecular mechanism. CG treatment significantly reduced HO-induced cell mortality and attenuated the production of lactate dehydrogenase and malondialdehyde in a dose-dependent manner.
