Iron Chelation Therapy Elicits Innate Immune Control of Metastatic Ovarian Cancer.

Cancer Discov

October 2024

Department of Obstetrics and Gynecology, Weill Cornell Medicine, New York, New York.

Iron accumulation in tumors contributes to disease progression and chemoresistance. Although targeting this process can influence various hallmarks of cancer, the immunomodulatory effects of iron chelation in the tumor microenvironment are unknown. Here, we report that treatment with deferiprone, an FDA-approved iron chelator, unleashes innate immune responses that restrain ovarian cancer.

Article Synopsis
  • * Recent research connects intramyocardial hemorrhage, which can occur when blood supply is restored post-MI, to a higher risk of developing CHF.
  • * The study indicates that iron from the hemorrhage triggers harmful processes in the heart tissue, causing fatty degeneration and negative remodeling of the heart, which could inform future treatment and management strategies for CHF following MI.

Diamond-Blackfan anaemia with iron overload: A serious issue.

Br J Haematol

October 2022

Pediatric Onco-Hematology, Stem Cell Transplantation and Cellular Therapy Division, Regina Margherita Children's Hospital, Azienda Ospedaliera-Universitaria Città della Salute e della Scienza, University of Torino, Torino, Italy.

Transfusion-dependent Diamond-Blackfan anaemia (DBA) patients rapidly develop iron overload and frequently experience cardiac complications. The report by Lecornec and colleagues offers useful details on indications and the management of deferiprone, a highly efficient chelator in removing excess cardiac iron but associated with a high risk of agranulocytosis in DBA patients. Commentary on: Lecornec et al.

Iron chelation promotes mitophagy through SENP3-mediated deSUMOylation of FIS1.

Autophagy

The selective clearance of mitochondria by mitophagy is an important quality control mechanism for maintaining mitochondrial and cellular health. Iron chelation, for example by the compound deferiprone (DFP), leads to a specific form of PINK1-PRKN/Parkin-independent mitophagy; however, the molecular mechanisms underlying this are poorly understood. In our recent paper, we examined the role of the deSUMOylating enzyme SENP3 in DFP-induced mitophagy.
