Background: Cortical excitability is elevated in Alzheimer's disease (AD). Transcranial magnetic stimulation-evoked responses on electromyography (EMG) and electroencephalography (EEG) have captured this increased excitability in motor brain regions. However, it is not yet known if increased excitability is also present in the parietal lobe or the extent to which excitability is related to cognition.

The proposed mechanisms underlying neuronal damage involve chronic low-grade inflammation, the production of reactive oxidative species, and altered brain glucose metabolism. In Alzheimer's disease, amyloid is thought to influence tau deposition and glucose metabolism.

PrecivityAD® incorporates plasma Aβ42/40, age and apoE proteotype into an algorithm that generates an amyloid probability score (APS) corresponding to the likelihood of a positive amyloid PET scan, defined as ≥25 centiloid (CL). A prototype PrecivityAD algorithm validated against amyloid PET status scored by visual read (VR) was used to improve screening efficiency in INVOKE-2, a phase 2 randomized, double-blind, placebo-controlled trial evaluating TREM2-activating antibody AL002 in early AD.

Method: A randomized, double-blind, placebo-controlled crossover design was used for this trial. Participants with MCI (n = 59) were sorted into one of two sequences: Sham-tVNS or tVNS-Sham.

Background: The Latarjet and other bony augmentation procedures are commonly used to treat anterior shoulder instability in the setting of significant glenoid bone loss. Although several fixation strategies have been reported, the biomechanical strength of these techniques remains poorly understood.
Purpose: To perform a systematic review of the biomechanical strength of glenoid bony augmentation procedures for anterior shoulder instability.
