Background: Human recombinant erythropoietin (rHuEPO) is often used in the treatment of diseases associated with a decreased production of red blood cells (RBC), such as chronic renal failure. rHuEPO is typically administered as an intravenous or subcutaneous (SC) injection every few days. The low minimum effective concentration (MEC) of rHuEPO, compared to the concentrations observed after standard doses, suggests that a low dose of the drug administered as a long-term infusion should be efficacious. This study aimed to compare the efficacy observed after a single subcutaneous administration of rHuEPO with that observed after a long-term infusion of rHuEPO via implanted osmotic pumps at a similar or lower dose.

Materials And Methods: In this study three rats received rHuEPO as a single SC injection at a dose of 1350 IU/kg, nine via osmotic pumps at a rate of 0.25, 0.5 and 1 IU/kg and at a total dose of 333 IU/kg, 667 IU/kg, 1333 IU/kg. Three rats served as a control group. The erythropoietin concentrations, RBC count and hemoglobin were measured.

Results: An increase in RBC count and hemoglobin was observed after SC infusion of rHuEPO. The baseline corrected area under the effect curve for hemoglobin and RBC count was more than 10-times higher for the SC infusion than for a single SC administration with a comparable dose.

Conclusions: This study demonstrates that administration of rHuEPO as a long-term infusion at a rate ensuring MEC allows to achieve a high efficacy of therapy using relatively small doses of the drug.

Background Aims: With novel therapies improving prognosis, the complications of multiple myeloma after multi-line treatment, particularly myelosuppression, have become a crucial determinant of long-term outcomes. Non-myeloablative allogeneic hematopoietic stem cell transplantation is a feasible option, but the transplant-related mortality rate remains high. Our study presents a relapsed/refractory multiple myeloma patient with a 9-year disease history.

Autoimmune nodopathy (AN) is a rare immune-mediated neuropathy characterized by autoantibodies against nodal or paranodal proteins. Patients with AN generally respond poorly to immunoglobulin therapy, and as a newly defined condition, there are currently no established treatment guidelines. Although rituximab shows potential as a therapeutic option, its high cost, limited availability, and the need for infusion monitoring hinder its use as a first-line treatment in many countries.

Pemphigus vulgaris Treated With Rituximab: A Case Report.

Cureus

November 2024

Dermatology, Hospital Dr. Nélio Mendonça, Serviço de Saúde da Região Autónoma da Madeira (SESARAM), Funchal, PRT.

This case report highlights the efficacy of rituximab (RTX), a monoclonal antibody that targets B-lymphocytes, in the treatment of severe pemphigus vulgaris (PV) that was unresponsive to multiple conventional therapies. A 44-year-old female presented with mucocutaneous lesions that had been progressing for 12 months, with a Pemphigus Disease and Area Index (PDAI) total activity score of 66, indicating severe disease. The patient received two infusions of RTX two weeks apart, without complications.

Enhancing Quasi-Solid-State Lithium-Metal Battery Performance: Multi-Interlayer, Melt-Infused Lithium and Lithiophilic Coating Strategies for Interfacial Stability in Li||VS-DSGNS-LATP|PEO-PVDF||NMC622-AlO Systems.

The development of quasi-solid-state lithium metal batteries (QSSLMBs) is hindered by inadequate interfacial contact, poor wettability between electrodes and quasi-solid-state electrolytes, and significant volume changes during long-term cycling, leading to safety risks and cataclysmic failures. Here, we report an innovative approach to enhance interfacial properties through the construction of QSSLMBs. A multilayer design integrates a microwave-synthesized LiAlTi(PO) (LATP) ceramic electrolyte, which is surface-coated with a lithiophilic conductive ink comprising VS and disulfonated functionalized graphene nanosheets (VS-DSGNS) using a low-cost nail-polish binder.

Patients with generalized myasthenia gravis (gMG) suffer from significant physical and social burdens. Although immunotherapies have been widely used for the treatment of gMG, some patients do not achieve or maintain remission. Recently, several molecular-targeting therapies of gMG, including the intravenous infusion of efgartigimod alfa (efgartigimod IV), a neonatal Fc receptor inhibitor, have been developed and are clinically used in Japan.
