Effective and safe vaccine adjuvants are needed to appropriately augment mucosal vaccine effects. Our previous study demonstrated that lipopolysaccharide (LPS) from Peyer's patch resident stimulated dendritic cells to promote the production of mucosal immunity-enhancing cytokines (e.g., IL-6 and BAFF), thus enhancing antigen-specific immune responses (including IgA production and Th17 responses) without excessive inflammation. Here, we chemically synthesized lipid A, the biologically active part of LPS, and examined its efficacy as a nasal vaccine adjuvant for the induction of protectively immunity against infection. Mice were nasally immunized with pneumococcal surface protein A (PspA) as a vaccine antigen for , together with lipid A. lipid A supported the generation of high levels of PspA-specific IgA and IgG responses through the augmentation of germinal center formation in the nasopharynx-associated lymphoid tissue and cervical lymph nodes (CLNs). Moreover, lipid A promoted PspA-specific CD4 Th17 responses in the CLNs and spleen. Furthermore, neutrophils were recruited to infection sites upon nasal infection and synchronized with the antigen-specific T and B cell responses, resulting in the protection against infection. Taken together, lipid A could be applied to the prospective adjuvant to enhance nasal vaccine efficacy by means of augmenting both the innate and acquired arms of mucosal immunity against respiratory bacterial infection.

Neisseria gonorrhoeae, which causes the sexually transmitted infection gonorrhea and Neisseria meningitidis, a leading cause of bacterial meningitis and septicemia, are closely related human-restricted pathogens that inhabit distinct primary mucosal niches. While successful vaccines against invasive meningococcal disease have been available for decades, the rapid rise in antibiotic resistance has led to an urgent need to develop an effective gonococcal vaccine. Several surface antigens are shared among these two pathogens, making cross-species protection an exciting prospect.

The objective was to determine the effects of injectable trace minerals (ITM, containing Se, Cu, Zn & Mn) administered at the time of primary intranasal (IN) modified-live virus (MLV) vaccination of young dairy calves on the serum neutralizing antibody (SNA) titers to Bovine herpes virus 1 (BHV1), Bovine respiratory syncytial virus (BRSV), and Bovine Parainfluenza type 3 virus (BPIV); cytokine expression in peripheral white blood cells, and BHV1-specific IgA titers in nasal secretions following the vaccination. A total of 60 calves (1 month old) were administered an IN MLV vaccine containing BHV1, BRSV, BPIV (Inforce 3) and randomly assigned to one of two experimental groups: ITM (n = 30; Multimin90, containing Se, Cu, Zn, and Mn) or SAL (n = 30; sterile saline). There was a consistent decay in virus-specific SNA titers in both groups.

Dry powders offer the potential to increase stability and reduce cold-chain requirements associated with the distribution of vaccines and other thermally sensitive products. The Alberta Idealized Nasal Inlet (AINI) is a representative geometry for characterization of nasal products that may prove useful in examining intranasal delivery of powders. Spray-dried trehalose powders were loaded at 10, 20, and 40 mg doses into active single-dose devices.
