Nonlinear optical microscopy allows for rapid high-resolution microscopy with image contrast generated from the intrinsic properties of the sample. Established modalities, such as multiphoton excited fluorescence and second/third-harmonic generation, can be combined with other nonlinear techniques, such as coherent Raman spectroscopy, which typically allow chemical imaging of a single resonant vibrational mode of a sample. Here, we utilize a single ultrafast laser source to obtain broadband coherent Raman spectra on a microscope, together with other nonlinear microscopy approaches on the same instrument. We demonstrate that the coherent Raman modality allows broadband measurement (>1000 cm), with high spectral resolution (<5 cm), with a rapid spectral acquisition rate (3-12 kHz). This enables Raman hyperspectral imaging of kilo-pixel images at >11 frames per second.

Broadband CARS is a coherent Raman scattering technique that provides access to the full biological vibrational spectrum within milliseconds, facilitating the recording of widefield hyperspectral Raman images. In this work, BCARS hyperspectral images of unstained cells from two different cell lines of immune lineage (T cell [Jurkat] and pDCs [CAL-1]) were recorded and analyzed using multivariate statistical algorithms in order to determine the spectral differences between the cells. A classifier was trained which could distinguish the known cells with a 97% out-of-bag accuracy.

A coherent concatenation of multiple solitary waves may lead to a stable infrared and visible broadband filament in a ceramic YAG polycrystal. This self-trapped soliton train is leveraged to implement self-referenced multiplex coherent anti-Stokes Raman scattering (SR-M-CARS) imaging. Simulations and experiments illustrating the filamentation process and the concatenation of focusing-defocusing cycles in ceramic and crystal YAG are presented.

Phenotype and genetic spectrum of six Indian patients with bestrophinopathy.

Taiwan J Ophthalmol

December 2024

Shri Bhagwan Mahavir Vitreoretinal Services, Medical Research Foundation, Sankara Nethralaya, Chennai, Tamil Nadu, India.

The aim of this study is to describe genotype and phenotype of patients with bestrophinopathy. The case records were reviewed retrospectively, findings of multimodal imaging such as color fundus photograph, optical coherence tomography (OCT), fundus autofluorescence, electrophysiological, and genetic tests were noted. Twelve eyes of six patients from distinct Indian families with molecular diagnosis were enrolled.

We recently demonstrated molecular plasmons in cyanine dyes for the conversion of photon energy into mechanical energy through a whole-molecule coherent vibronic-driven-action. Here we present a model, a molecular plasmon analogue of molecular orbital theory and of plasmon hybridization in metal nanostructures. This model describes that molecular plasmons can be obtained from the combination or hybridization of elementary molecular fragments, resulting in molecules with hybridized plasmon resonances in the electromagnetic spectrum.

Coherent Anti-Stokes Hyper-Raman Spectroscopy.

Nat Commun

Coherent Raman scattering spectroscopies have been established as a powerful tool for investigating molecular systems with high chemical specificity. The existing coherent Raman scattering techniques detect only Raman active modes, which are a part of the whole molecular vibrations. Here, we report the first observation of coherent anti-Stokes hyper-Raman scattering (CAHRS) spectroscopy, which allows measuring hyper-Raman active vibrations at high speed.
