Background: Peak-width of skeletonized mean diffusivity (PSMD) is an emerging biomarker of cerebral small vessel disease (cSVD)-related vascular contributions to cognitive impairment and dementia (VCID). Higher PSMD values reflect greater white matter microstructural damage, and prior research has related PSMD to sporadic and monogenic forms of cSVD and worse cognitive function. Therefore, we proposed PSMD as a risk stratification biomarker for VCID.

View Article and Find Full Text PDFBackground: Cardiovascular-kidney-metabolic (CKM) health, a term recently defined by the American Heart Association, encompasses the interplay among metabolic, chronic kidney, and cardiovascular risk factors. We aimed to investigate the predictive significance of CKM disorders with the risk of cognitive decline and Alzheimer's disease (AD) and AD-related dementia (ADRD) mortality in a multiethnic population.
Method: We analyzed a cohort of 6,440 adults aged 45-84 who participated in the Multiethnic Study of Atherosclerosis, with a baseline survey conducted in 2000-2002, and were followed through to December 2015.
Background: In older adults, greater amyloid (Aβ) and tau positron emission tomography (PET) binding is associated with cognitive decline and dementia. However, the association of early amyloid and tau PET accumulation with cognition at midlife remains unclear. The goal of the current study was to evaluate the associations of Aβ and tau PET with cognition in a predominately middle-aged community-based cohort, as well as to examine the factors that may modify these associations.

Background: The unique lesion of chronic traumatic encephalopathy (CTE) is the perivascular deposition of hyperphosphorylated tau at the depth of the cortical sulci. The distribution and molecular composition of p-tau is distinct from Alzheimer's disease (AD), but differential diagnostic challenges remain. Understanding disease differences in regional density of p-tau will inform differential diagnosis and interpretation of in vivo biomarkers.
