Combined nitrogen and drought stress leads to overlapping and unique proteomic responses in potato.

Planta

February 2023

Department of Woody Plant and Propagation Physiology, Institute of Horticultural Production Systems, Leibniz University Hannover, Herrenhäuser Straße 2, 30419, Hannover, Germany.

Nitrogen deficient and drought-tolerant or sensitive potatoes differ in proteomic responses under combined (NWD) and individual stresses. The sensitive genotype 'Kiebitz' exhibits a higher abundance of proteases under NWD. Abiotic stresses such as N deficiency and drought affect the yield of Solanum tuberosum L.

Semi-Thermal Asymmetric Reverse PCR (STARP) Genotyping.

Methods Mol Biol

February 2023

Department of Plant Sciences, Quaid-i-Azam University, Islamabad, Pakistan.

PCR-based individual Single nucleotide polymorphism (SNP) genotyping methods are preferred due to their flexibility, high-throughput, and improved accuracy. Semi-thermal asymmetric reverse PCR (STARP) is one of the SNP genotyping methods developed to reduce operational cost with improved platform compatibility. STARP is a unique method which can be used either as a gel-free SNP genotyping by detection of fluorescent signals or polyacrylamide gel-based size separation.

National Key Facility for Crop Gene Resources and Genetic Improvement, Institute of Crop Sciences, Chinese Academy of Agricultural Sciences, Beijing, China.

High-throughput genotyping for functional markers offers an excellent opportunity to effectively practice marker-assisted selection (MAS) while breeding cultivars. We developed kompetitive allele-specific PCR (KASP) assays for genes conferring drought tolerance in common wheat ( L.).

Interactions of organisms with their environment are complex and environmental regulation at different levels of biological organization is often nonlinear. Therefore, the genotype to phenotype continuum requires study at multiple levels of organization. While studies of transcriptome regulation are now common for many species, quantitative studies of environmental effects on proteomes are needed.

Factor V Leiden (FV ) is a missense mutation of 1691 position (G1691A) in exon 10 of FV gene, and being a genetic risk for venous thrombosis. Currently, there are several PCR-based methods for detecting FV mutation; however, these methods have disadvantages such as time-consuming, cumbersome steps and potentially hazardous gels. The aims of present study were to develop a simple, time-saving, accurate, and gel-free method, called amplification refractory mutation system (ARMS) TaqMan real-time PCR, for detecting FV mutation.
