Side-chain conformers to allow conversion from normal to isoaspartate in age-related proteins and peptides.

Dβ (or D-iso)- and Lβ- (or iso)- aspartyl (Asp) residues are accumulated in aged lens crystallins and amyloid beta (Aβ) proteins, respectively, as a result of spontaneous, nonenzymatic isomerization of normal Lα-Asp. To explore why such uncommon Asp isomers are accumulated, the stability of Lα-, Lβ-, and Dβ-Asp was compared in view of the staggered side-chain conformers. By using cylindrin (KVKVLGDVIEV) from αB-crystallin and Aβ17-25 (LVFFAED)VG) containing Asp isomers, the vicinal spin-spin coupling constants of Asp H-H and H-H were quantified by high-resolution solution H NMR. It was found that the trans conformer was extremely preferred in Dβ-Asp7 side-chain of cylindrin. In Aβ17-25, the side chain of Lβ-Asp23 was likely to adopt trans conformer, while gauche conformers were rather rich in Lα-Asp23. In gauche conformers, the close distance between Asp carboxylate carbon (C) and backbone nitrogen (N) next to Asp is advantageous to the intramolecular cyclization to form succinimide intermediate, followed by the conversion from α- to β-Asp. The cyclization is limited in the trans conformer because of the long distance between C and N, to keep Dβ- or Lβ-Asp stable. This would be the reason for the site specificity of Asp isomerization in proteins. The higher population of trans conformer in Asp side chain, the less isomerization of Asp as shown as Asp76 in αA-crystallin. The stability and less reactivity of normal Asp and its isomers are the potential factors to determine whether or not the abnormal accumulation is permitted in aged crystallins and Aβ.

2-Deoxy-β-glycosides are essential components of natural products and pharmaceuticals; however, the corresponding 2-deoxy-β-glycosidic bonds are challenging to chemically construct. Herein, we describe an efficient catalytic protocol for synthesizing 2-deoxy-β-glycosides via either IPrAuNTf-catalyzed activation of a unique 1,2--positioned C2--propargyl xanthate (OSPX) leaving group or (PhO)PAuNTf-catalyzed activation of a 1,2--C2--alkynylbenzoate (OABz) substituent of the corresponding thioglycosides. These activation processes trigger 1,2-alkyl/arylthio-migration glycosylation, enabling the synthesis of structurally diverse 2-deoxy-β-glycosides under mild reaction conditions.

The structural stability of the energetic material 2,2',4,4',6,6'-hexanitrostilbene (-HNS) under high pressure is critical for optimizing its detonation performance and low sensitivity. Loz1 is a zinc-responsive transcription factor in fission yeast that maintains cellular zinc homeostasis by repressing the expression of genes required for zinc uptake in high zinc conditions. Previous deletion analysis of Loz1 found a region containing two tandem CH zinc-fingers and an upstream "accessory domain" rich in histidine, lysine, and arginine residues to be sufficient for zinc-dependent DNA binding and gene repression. Here we report unexpected biophysical properties of this pair of seemingly classical CH zinc fingers.

-β, β-β' trifused porphyrins incorporating two distinct active methylene groups (MN = malononitrile and IND = 1,3-indanedione) and their corresponding metal complexes with Cu(II) and Zn(II) have been synthesized with good to excellent yields and characterized by various spectroscopic techniques and spectrometric methods. Single crystal X-ray analysis of the Zn(II) complex ZnTFPMB(MN) (where TFP = trifused porphyrin and MB = mono benzo) revealed a nonplanar 'armchair' type conformation with a twist angle of 24.10°.

A multiomic study of the structural characteristics of type A and B influenza viruses by means of highly spectrally resolved Raman spectroscopy is presented. Three virus strains, A H1N1, A H3N2, and B98, were selected because of their known structural variety and because they have co-circulated with variable relative prevalence within the human population since the re-emergence of the H1N1 subtype in 1977. Raman signatures of protein side chains tyrosine, tryptophan, and histidine revealed unequivocal and consistent differences for pH characteristics at the virion surface, while different conformations of two C-S bond configurations in and methionine rotamers provided distinct low-wavenumber fingerprints for different virus lineages/subtypes.
