The polyhistidine tag (His-tag) is one of the most popular protein tags used in the life sciences. Traditionally, the detection of His-tagged proteins relies on immunoblotting with anti-His antibodies. This approach is laborious for certain applications, such as protein purification, where time and simplicity are critical. The His-tag can also be directly detected by metal ion-loaded nickel-nitrilotriacetic acid-based chelator heads conjugated to fluorophores, which is a convenient alternative method to immunoblotting. Typically, such chelator heads are conjugated to either green or red fluorophores, the detection of which requires specialized excitation sources and detection systems. Here, we demonstrate that post-run staining is ideal for His-tag detection by metal ion-loaded and fluorescently labeled chelator heads in PAGE and blot membranes. Additionally, by comparing the performances of different chelator heads, we show how differences in microscopic affinity constants translate to macroscopic differences in the detection limits in environments with limited diffusion, such as PAGE. On the basis of these results, we devise a simple approach, called UVHis-PAGE, that uses metal ion-loaded and fluorescently labeled chelator heads to detect His-tagged proteins in PAGE and blot membranes. Our method uses a UV transilluminator as an excitation source, and the results can be visually inspected by the naked eye.

We present a case series consisting of three female patients in their 30s with presumed autoimmune uveitis resembling Cogan's syndrome following caesarean sections (C-sections) with severe intraoperative bleeding and the use of chitosan-tamponade, exhibiting a combination of varying ocular and auditory symptoms postoperatively. Our patients displayed a range of inflammatory ocular changes, including stromal keratitis, panuveitis, retinal infiltrates, haemorrhages, optic disc swelling, and intraretinal and subretinal fluid, along with otalgia and hearing loss, consistent with typical and atypical Cogan's syndrome. Treatment involved systemic corticosteroids, resulting in variable outcomes.

Objective: Our study aimed to investigate the ototoxicity associated with the iron chelator deferasirox in patients with β-thalassemia major, who were receiving regular transfusion therapy, along with evaluating the data on audiological tests using appropriate statistical tests.
Methods: A cross-sectional observational study was conducted on 100 transfusion-dependent β-thalassemia major patients on oral iron chelating agent-deferasirox. Pure tone audiometry (PTA) and distortion product otoacoustic emissions (DPOAE) was carried out in all patients to assess the auditory side effects of the drug.

Infectious acute pneumonia caused by bacteria has been a great challenge to human health for long time, and the rapid clearance of aerosolized antibiotics in the lungs restricts their clinical application. The development of nanoformulations with facile preparation and mucoadhesive properties for the pulmonary delivery of antibiotics is thus significant for the treatment of infectious acute pneumonia. In this study, FDA (Food and Drug Administration)-approved tannic acid (TA) was used to construct mucoadhesive nanoformulations through the facile coating of chitosan (CS) to achieve long-lasting anti-infection effects against infectious acute pneumonia.
