Erwinia amylovora is the causal agent of fire blight, a devastating disease of apples and pears. A previous study revealed that an E. amylovora uracil auxotroph was still virulent and can cause disease, suggesting that uracil can be obtained from the host environment. The E. amylovora genome contains a locus encoding for a uracil transporter belonging to the nucleobase cation symporter 2 family, displaying a high level of amino acid sequence similarity to the Escherichia coli UraA. Expression of E. amylovora UraA in nucleobase transporter-deficient E. coli strains, coupled with radiolabeled uptake studies reveal that E. amylovora UraA is a high affinity uracil transporter with a K of 0.57 μM. Both E. coli and E. amylovora carrying extra copies of E. amylovora UraA are sensitive to growth on the toxic analog 5-fluorouracil. An E. amylovora ΔuraA::Cam mutant is still able to grow and cause disease symptoms on immature pears and apples.

Chemotherapy resistance is a great challenge in the treatment of gastric cancer (GC), so it is urgent to explore the prognostic markers of chemoresistance. PUF60 (Poly (U)-binding splicing factor 60) is a nucleic acid-binding protein that has been shown to regulate transcription and link to tumorigenesis in various cancers. However, its biological role and function in chemotherapy resistance of GC is unclear.

Gastric cancer (GC) ranks as the fifth leading cause of cancer mortality, with cancer stem cells (CSCs) playing a critical role in tumor progression and resistance to chemotherapy. Conventional chemotherapy often fails to effectively target these stem cells. BATF2, a tumor suppressor, is known for its role in gastric cancer, but its influence on cancer stem cell-like properties and chemotherapy response remains unclear.

Although combining 5-fluorouracil (5-FU) and Interferon-beta (IFNb) improves response rates in Hepatocellular Carcinoma (HCC), the outcomes remain suboptimal. This study investigates the feasibility of using highly transfected Mesenchymal Stem Cells (MSCs) to deliver a chemotherapeutic (5-FU) and an immunomodulator (IFNb) for localized HCC treatment. Considering the crucial role of cold-chain transportation in off-the-shelf allogeneic therapy, the study also assesses the quality and efficacy of frozen-thawed engineered MSCs, simulating a multisite study process.

Effective drug release is of utmost importance in the medical field for treating various diseases, particularly cancer. Nanocomposite hydrogels remain the best materials for enhancing the bioavailability and therapeutic levels of drugs as they enable sustained, targeted, or controlled drug release. In this work, a nanocomposite hydrogel containing locust bean gum (LBG), poly(4-acryloylmorpholine) (PAcM), and silver nanoparticles (SN) has been made using an eco-friendly microwave (MW)-assisted method and characterized by various advanced techniques.

Unlabelled: is an opportunistic pathogen that can salvage nucleobases from the environment to conserve nutrients that would otherwise be spent on nucleotide biosynthesis. However, little is known regarding the substrate specificity of the 13 putative nucleobase transporters in . Here, using a combination of genetic and chemical approaches, we report substrate identifications for 10 putative nucleobase transporters in .
