The outbreak of SARS-CoV-2 is posing serious global public health problems. Facing the emergence of this pandemic, we established a portable microfluidic immunoassay system for easy-to-use, sensitive, rapid (<15 min), multiple, and on-site detection of IgG/IgM/Antigen of SARS-CoV-2 simultaneously. This integrated method was successfully applied for detecting SARS-CoV-2 IgM and IgG antibodies in clinical human serum as well as SARS-CoV-2 antigen in pharyngeal swabs from 26 patients with COVID-19 infection and 28 uninfected people. The assay demonstrated high sensitivity and specificity, which is promising for the diagnosis and monitoring as well as control of SARS-CoV-2 worldwide.

Background: Plasmonic core-shell nanostructures with embedded internal markers used as Raman probes have attracted great attention in surface-enhanced Raman scattering (SERS) immunoassay for cancer biomarkers due to their excellent uniform enhancement. However, current core-shell nanostructures typically exhibit a spherical shape and are coated with a gold shell, resulting in constrained local field enhancement.
Results: In this work, we prepared a core-shell AuNR@BDT@Ag structure by depositing silver on the surface of Raman reporter-modified gold nanorods (AuNR).

Rapid and sensitive detection of Epstein-Barr virus cell-free DNA (EBV cfDNA) is crucial for early diagnosis and monitoring of nasopharyngeal carcinoma (NPC), but accessibility to screening is limited by complicated and costly conventional DNA isolation and purification approaches. Here, a fully integrated ion concentration polarization (ICP)-enriched and nanozyme-catalyzed lateral flow assay (ICP-cLFA) is developed, enabling total analysis of EBV cfDNA in whole blood samples, with DNA isolation, pre-concentration, and amplification performed on a microfluidic chip, consequently providing the signal readout within 75 min. Specifically, ICP preconcentration and amplification steps, together with target recognition catalyzed by a platinum-decorated mesoporous gold nanosphere (MGNS@Pt) nanozyme, result in an ultralow detection limit of 4 aM in standard cfDNA samples and 100 aM in whole blood from NPC-bearing rats.

Methods: This study aimed to evaluate serum levels of cholesterol, HDL, and HDL subfractions (HDL-SUB) to define their role in the pathogenesis of MIS-C and their potential use as biomarkers of this condition.
