We have previously proposed that the reciprocal hindlimb scratching (RHS) of mice elicited by intrathecal injection of muscarinic agents is mediated by M1 muscarinic receptors. In this study, benzhexol potently inhibited pilocarpine-induced RHS (ID50 = 0.5 ng), methoctramine did not fully block RHS and RS 86 neither produced nor blocked RHS. These data (1) differentiate the three muscarinic agents using this in vivo endpoint, (2) substantiate binding studies characterizing benzhexol and methoctramine as putative M2 and M2 antagonists, respectively, and (3) further support using RHS for studying muscarinic agents.

Objective: Organophosphorus Nerve Agent, VX [(O-Ethyl S-diisopropylaminomethyl) methylphosphonothioate] compound interferes with acetylcholine signaling by targeting the AChE enzyme. Studies suggest that in nerve agents poisoning, non-cholinergic effects are also responsible for damage in peripheral tissues including long term damage in brain. Present study reports cholinergic and non-cholinergic effects of VX poisoning and their prevention by use of N-acetylcysteine (NAC) in addition to conventional antidotes atropine sulphate and 2-PAM chloride as an antioxidant.

Anticholinergic drugs are widely used for very common diseases such as Parkinson's disease, depression or allergies, but it has been observed in previous studies that high anticholinergic burden can lead to physical and cognitive dysfunctions in polymedicated elderly people. The objective of this project is to review these hypotheses and study the possible relationship between high anticholinergic burden and cognitive and physical dysfunctions.In terms of methodology, a bibliographic review has been carried out of the information that exists to date on the hypothesis that high anticholinergic burden can lead to both physical and cognitive dysfunctions in elderly people taking multiple medications, and a cross-sectional study has been designed that included patients over 70 years and taking multiple medications to calculate the anticholinergic burden of their medication and determine whether high levels of it could have any influence on the deterioration of the patients.

Cytochrome P450 (CYP) 3A4 plays a major role in drug metabolism. Its activity could be determined by non-invasive and cost-effective assays, such as breath analysis, for the personalised monitoring of drug response. For the first time, we identify an isotopically unlabelled CYP3A4 substrate, tolterodine that leads to the formation of a non-toxic volatile metabolite, acetone, which could potentially be applied to monitor CYP3A4 activity in humans.
