2-(Arylamino)-4,6-di-tert-butylphenols containing 4-substituted phenyl groups (RapH2) react with oxobis(ethylene glycolato)osmium(vi) in acetone to give square pyramidal bis(amidophenoxide)oxoosmium(vi) complexes. A mono-amidophenoxide complex is observed as an intermediate in these reactions. Reactions in dichloromethane yield the diolate (Hap)2Os(OCH2CH2O). Both the glycolate and oxo complex are converted to the corresponding cis-dichloride complex on treatment with chlorotrimethylsilane. The novel bis(aminophenol) ligand EganH4, containing an ethylene glycol dianthranilate bridge, forms the chelated bis(amidophenoxide) complex (Egan)OsO, where the two nitrogen atoms of the tetradentate ligand bind in the trans positions of the square pyramid. Structural and spectroscopic features of the complexes are described well by an osmium(vi)-amidophenoxide formulation, with the amount of π donation from ligand to metal increasing markedly as the co-ligands change from oxo to diolate to dichloride. In the oxo-bis(amidophenoxides), the symmetry of the ligand π orbitals results in only one effective π donor interaction, splitting the energy of the two osmium-oxo π* orbitals and rendering the osmium-oxo bonding appreciably anisotropic.

ConspectusMetal oxo (M═O) and nitrido (M≡N) complexes are two important classes of high-valent transition metal complexes. The use of M═O as oxidants in chemical and biological systems has been extensively investigated. Nature makes use of M═O in enzymes such as cytochrome to oxidize a variety of substrates.

A bare lead atom is a σ-donor ligand capable of linearly bonding and stabilizing two units of a classical polyhydride complex, with a high-valent metal center. As a proof of concept, we have prepared and characterized the μ-tetrylide complex (PPr)HOs═Pb═OsH(PPr) in the reaction of OsH(PPr) with Pb{N(SiMe)}. Although the Pb-Os bonds exhibit electrostatic interaction, the main orbital interactions result from two dative σ bonds from the lead atom to the osmium centers.

Interest in the third-row transition metal osmium and its compounds as potential anticancer agents has grown in recent years. Here, we synthesized the osmium(VI) nitrido complex (tpm = [5-(Thien-2-yl)-1H-pyrazol-3-yl]methanol), which exhibited a greater inhibitory effect on the cell viabilities of the cervical, ovarian, and breast cancer cell lines compared with cisplatin. Proteomics analysis revealed that modulates the expression of protein-transportation-associated, DNA-metabolism-associated, and oxidative-stress-associated proteins in HepG2 cells.

Article Synopsis
  • Resonant X-ray emission spectroscopy (RXES) is an advanced technique developed over the past decade to analyze the chemical states of metal complexes and observe chemical reactions.
  • The study focused on using RXES to track how osmium(VI) nitrido complexes convert to osmium(III) ammine states when reduced by glutathione, a significant biological reducing agent.
  • The RXES results align with previous theoretical predictions and suggest the formation of osmium(IV) intermediates, supporting a pathway that may enhance the anticancer effects of these metal complexes when reduced inside cells.

The high stability of the experimentally known homoleptic 1-norbornyl derivative (nor)Fe of iron in the unusual +4 oxidation state is a consequence of the high reaction barriers of the singlet or triplet potential surfaces constrained by the global dispersion attraction and the great steric demands of the norbornyl groups. The much more limited stability of the corresponding cyclohexyl derivative (cx)Fe may result from the conical intersection between the singlet potential surface and the quintet spin potential surface arising from the weaker dispersion attraction and the reduced steric effect of the cyclohexyl groups relative to the 1-norbornyl groups. In contrast, the high stability of the likewise experimentally known (cx)M (M = Ru or Os) structures results from the larger ligand field splitting (Δ) of the d-orbital energies for the second and third-row transition metals ruthenium and osmium relative to that of the first-row transition metal iron.
