Glutamic acid decarboxylase 65-kilodalton isoform (GAD65) antibodies have been associated with multiple nonneurological and neurological syndromes including autoimmune epilepsy (AE). Although immunotherapy remains the cornerstone for the treatment of AE, those with GAD65 Ab-associated AE (GAD65-AE) remain refractory to immunotherapy and antiseizure medication (ASM). Outcomes of epilepsy surgery in this patient population have also been unsatisfactory. The role of neuromodulation therapy, particularly direct brain-responsive neurostimulation therapy, has not been previously examined in GAD65-AE. Here, we describe four consecutive patients with refractory GAD-65-associated temporal lobe epilepsy (GAD65-TLE) receiving bilateral hippocampal RNS System treatment. The RNS System treatment was well tolerated and effective in this study cohort. Three patients had a >50% clinical seizure reduction, and one patient became clinically seizure-free following resective surgery informed by the RNS System data with continued RNS System treatment. In all four of our patients, the long-term ambulatory data provided by the RNS System allowed us to gain objective insights on electrographic seizure lateralization, patterns, and burden as well as guided immunotherapy and ASM optimization. Our results suggest the potential utility of the RNS System in the management of ASM intractable GAD65-AE.

OH-mediated advanced oxidation processes (AOPs) are widely used in wastewater treatment and drinking water purification. Recently, an increasing number of studies have indicated that common inorganic nitrogen ions can efficiently generate •OH under UV irradiation, demonstrating strong performance in the degradation of various contaminants. Conversely, the presence of inorganic nitrogen ions in UV or other oxidation processes dramatically increases the yield of toxic nitro (so)-aromatic products and the formation potential of nitrogenous disinfection by-products with high genotoxicity and cytotoxicity.

Monitoring reactive nitrogen species (RNS) in complex biological media is essential for evaluating the health status of living organisms; however, biofouling on the sensor surface restricts its applications. To overcome this issue, we developed an antifouling electrochemical sensing platform using copper-platinum bimetallic nanoparticles/N-doped biomass porous carbon fibres (Cu-PtNPs/N-BCF) for directly detecting peroxynitrite anion (ONOO), a major type of RNS. Cyclic voltammetry measurements demonstrated that the Cu-PtNPs/N-BCF-2 nanocomposite, synthesised at a molar ratio of 1:1 between Co and Zn, exhibited exceptional electrocatalytic activity for ONOO oxidation.

Although immunotherapy has revolutionized clinical cancer treatment, the efficacy is limited due to the lack of tumor-associated antigens (TAAs) and the presence of compensatory immune checkpoints. To overcome the deficiency, a nano-system loaded with ozone and CD47 inhibitor RRx-001 is designed and synthesized. Upon irradiation, reactive oxygen species (ROS) generated from ozone reacts with nitric oxide (NO) metabolized from RRx-001 to form reactive nitrogen species (RNS), which presents a much stronger cell-killing ability than ROS.

Background: Given the emotional and physical demands of nursing, especially in recent years, exploring ways that hope-inducing and resilience-building models can support professional practice is vital to the current and future nursing workforce.
Methods: An anonymous survey consisting of demographic questions, the Adult Hope Scale, Connor-Davidson Resilience Scale, and the Conditions for Work Effectiveness Questionnaire II was offered to 1450 RNs in a nonprofit community-based healthcare system for volunteer participation.
