The human body is able to process and transport a complex variety of carbohydrates, unlocking their nutritional value as energy source or as important building block. The endogenous glycosyl hydrolases (glycosidases) and glycosyl transporter proteins located in the enterocytes of the small intestine play a crucial role in this process and digest and/or transport nutritional sugars based on their structural features. It is for these reasons that glycosidases and glycosyl transporters are interesting therapeutic targets to combat sugar related diseases (such as diabetes) or to improve drug delivery. In this review we provide a detailed overview focused on the molecular structure of the substrates involved as a solid base to start from and to fuel research in the area of therapeutics and diagnostics.

Phoronida is a small group of marine animals, most of which are characterized by a long larval period and complex metamorphosis. As a result of metamorphosis, their body changes so much that their true anterior and posterior ends are very close to each other, and the intestine becomes long and U-shaped. Using histology and electron microscopy, we have shown that the elongation and change in shape of the digestive tract that occurs during metamorphosis in Phoronopsis harmeri larvae is accompanied by the formation of new parts and changes in ultrastructure.

The small intestine is the longest segment of the gastrointestinal (GI) tract, but cancers in the small intestine are infrequent. The duodenojejunal (DJ) flexure is an uncommon site for tumors, and those located in these sites are difficult to identify and manage properly. Their rarity, along with ambiguous symptoms that can be readily misattributed to milder conditions, results in a delayed diagnosis when the tumors have significantly advanced.

Total proctocolectomy with ileal pouch anal anastomosis is the standard of care for patients with severe ulcerative colitis. We generated a cell-type-resolved transcriptional and epigenetic atlas of ileal pouches using scRNA-seq and scATAC-seq data from paired biopsy samples of the ileal pouch and the ileal segment above the pouch (pre-pouch) from patients (male=4, female=2), and paired biopsies of the terminal ileum and ascending colon from healthy individuals (male=3, female=3) serving as reference. Our study finds an additional population of absorptive and secretory epithelial cells within the pouch but not the pre-pouch.
