Process characterization is necessary in the biopharmaceutical industry, leading to concepts such as design of experiments (DoE) in combination with process modeling. However, these methods still have shortcomings, including large numbers of required experiments. The concept of intensified design of experiments (iDoE) is proposed, that is, intra-experimental shifts of critical process parameters (CPP) that combine with hybrid modeling to more rapidly screen a particular design space. To demonstrate these advantages, a comprehensive experimental design of Escherichia coli (E. coli) fed-batch cultivations (20 L) producing recombinant human superoxide dismutase is presented. The accuracy of hybrid models trained on iDoE and on a fractional-factorial design is evaluated, without intra-experimental shifts, to simultaneously predict the biomass concentration and product titer of the full-factorial design. The hybrid model trained on data from the iDoE describes the biomass and product at each time point for the full-factorial design with high and adequate accuracy. The fractional-factorial hybrid model demonstrates inferior accuracy and precision compared to the intensified approach. Moreover, the intensified hybrid model only required one-third of the data for model training compared to the full-factorial description, resulting in a reduced experimental effort of >66%. Thus, this combinatorial approach has the potential to accelerate bioprocess characterization.

Background: Rural School Support Strategies (RS3) is a bundle of implementation supports (including training, technical assistance, and a virtual learning collaborative) designed for the scale-up of universal prevention initiatives. This study addresses mechanisms of action, exploring whether positive effects of RS3 on implementation fidelity are attributable to improvements in functioning of school implementation teams, and increases in organizational readiness.
Methods: Data are from a cluster-randomized hybrid Type 3 implementation-effectiveness trial of RS3 among rural Idaho schools implementing Positive Behavioral Interventions and Supports (PBIS).

In this study, two novel hybrid intelligent models were developed to evaluate the short-term rockburst using the random forest (RF) method and two meta-heuristic algorithms, whale optimization algorithm (WOA) and coati optimization algorithm (COA), for hyperparameter tuning. Real-time predictive models of this phenomenon were created using a database comprising 93 case histories, taking into account various microseismic parameters. The results indicated that the WOA achieved the highest overall performance in hyperparameter tuning for the RF model, outperforming the COA.

Reactive oxygen species (ROS) is promising in cancer therapy by accelerating tumor cell death, whose therapeutic efficacy, however, is greatly limited by the hypoxia in the tumor microenvironment (TME) and the antioxidant defense. Amplification of oxidative stress has been successfully employed for tumor therapy, but the interactions between cancer cells and the other factors of TME usually lead to inadequate tumor treatments. To tackle this issue, we develop a pH/redox dual-responsive nanomedicine based on the remodeling of cancer-associated fibroblasts (CAFs) for multi-pronged amplification of ROS (ZnPP@FQOS).
