Unlabelled: Rheumatoid arthritis (RA) is among the most prevalent forms of autoimmunity. Gentiopicroside (Gent) is an iridoid glucoside derived from the which is used in traditional Chinese medicine to treat RA. The present study was designed to explore the ability of Gent to combat RA and to explore the molecular basis for such anti-RA activity both using tumor necrosis factor alpha (TNF-α)-stimulated human RA fibroblast-like synoviocytes (RA-FLS) and using a rat adjuvant-induced arthritis (AIA) model. We found that Gent was able to significantly reduce the swelling of joints and arthritic index scores, with corresponding reductions in synovial inflammatory cell infiltration, synovial hyperplasia, and bone erosion in treated AIA rats. Importantly, Gent 200 mg/kg reduced thymus index in AIA rats, but had no effect on spleen index and body weight, it revealed that Gent was relatively safe at the dose we chose. We further found that Gent was able to suppress the TNF-α-induced proliferation and migration of RA-FLS cells. This suppression was attributed to the ability of Gent to block NOD-like receptor protein 3 (NLRP3), apoptosis-associated speck-like protein containing a CARD (ASC), and caspase-1, thereby disrupting the activation of the NLRP3 inflammasome. Consistent with such suppression, Gent led to a significant decrease in IL-1β secretion by treated cells. Furthermore, this reduction in NLRP3 inflammasome activation was also associated with decreases in the activation of nuclear factor (NF-κB), the production of reactive oxygen species (ROS), and the expression of inflammatory IL-6. Together these findings indicate that Gent can suppress the ROS-NF-κB-NLRP3 axis to alleviate RA symptoms.

Decoding ruminative reflection in healthy individuals: The role of triple network connectivity.

Int J Clin Health Psychol

October 2024

Key Laboratory of Cognition and Personality, Faculty of Psychology, Southwest University, Chongqing, China.

Ruminative reflection has been linked to enhanced executive control in processing internally represented emotional information, suggesting it may serve as an adaptive strategy for emotion regulation. Investigating the neural substrates of reflection can deepen our understanding of its adaptive properties. This study used network-based statistic (NBS)-Predict methodology to identify resting state functional connectivity (FC)-based predictors of ruminative reflection in a healthy sample.

This study investigated the effects of different protein sources on feed intake, nutrient, and energy utilization, growth performance, and enteric methane (CH4) emissions in growing beef cattle, also evaluated against a pasture-based diet. Thirty-two Holstein × Angus growing beef were allocated to four dietary treatments: a total mixed ration (TMR) including solvent-extracted soybean meal as the main protein source (SB; n = 8), TMR with local brewers' spent grains (BSG; n = 8), TMR with local field beans (BNS; n = 8), and a diet consisting solely of fresh-cut Italian ryegrass (GRA; n = 8). Every four weeks, animals were moved to digestibility stalls within respiration chambers to measure nutrient intakes, energy and nitrogen (N) utilization, and enteric CH4 emissions.

Purpose: Two randomized clinical trials (STOMP and ORIOLE) demonstrated that stereotactic ablative radiotherapy (SABR) can prolong ADT-free survival or progression-free survival (PFS) in patients with metachronous oligometastatic prostate cancer (omCSPC) patients. While most omCSPC patients have a more modest delay in progression, a small subset achieves a durable response following SABR. We investigated the prognostic and predictive value of circulating PSMA-positive extracellular vesicles (PSMA+EV) and prostate specific antigen (PSA) in a biomarker correlative study using blood samples from three independent patient cohorts.

Inflammatory bowel diseases cause chronic intestinal inflammation, including Crohn's disease (CD) and ulcerative colitis (UC). Prostaglandin E-major urinary metabolite (PGE-MUM) is a urine biomarker for disease activity in IBD. This study evaluated PGE-MUM performance for predicting an active disease in patients with CD and UC.

Volumetric bioprinting of the osteoid niche.

Biofabrication

January 2025

Volumetric bioprinting has revolutionized the field of biofabrication by enabling the creation of cubic centimeter-scale living constructs at faster printing times (in the order of seconds). However, a key challenge remains: developing a wider variety of available osteogenic bioinks that allow osteogenic maturation of the encapsulated cells within the construct. Herein, the bioink exploiting a step-growth mechanism (norbornene-norbornene functionalized gelatin in combination with thiolated gelatin - GelNBNBSH) outperformed the bioink exploiting a chain-growth mechanism (gelatin methacryloyl - GelMA), as the necessary photo-initiator concentration was three times lower combined with a more than 50 % reduction in required light exposure dose resulting in an improved positive and negative resolution.
