Early childhood caries (ECC) is a severe manifestation of carious pathology with rapid and disruptive progression. The ECC microbiota includes a wide variety of bacterial species, among which is an anaerobic newly named species, a previously unidentified . Our aim was to provide the first ultrastructural characterization of and its biofilm by scanning electron microscopy (SEM) using a protocol that faithfully preserved the biofilm architecture and allowed an investigation at very high magnifications (order of nanometers) and with the appropriate resolution. To accomplish this task, we analyzed ' biofilm by conventional SEM and VP-SEM protocols, in addition, we developed an original procedure, named OsO-RR-TA-IL, which avoids dehydration, drying and sputter coating. This innovative protocol allowed high-resolution and high-magnification imaging (from 10000× to 35000×) in high-vacuum and high-voltage conditions. After comparing three methods, we chose OsO-RR-TA-IL to investigate . It appeared as a fusiform elongated bacterium, without surface specialization, arranged in clusters and submerged in a rich biofilm matrix, which showed a well-developed micro-canalicular system. Our results provide the basis for the development of innovative strategies to quantify the effects of different treatments, in order to establish the best option to counteract ECC in pediatric patients.

Crystal-violet staining, microscopy with image analysis, and quantitative PCR (qPCR) were compared to examine biofilm dynamics. Biofilms of 30 polycultures comprising 15 bacterial species were monitored for 14 days. Collectively, qPCR (representing population) revealed a different growth pattern compared to staining (biomass) and microscopy (colonization): biomass and colonization gradually increased over time, whereas population increased rapidly for the first seven days and leveled off.

Propolis is a resinous material collected by different bee species from various plant exudates and used to seal holes in honeycombs, smoothen the internal walls, embalm intruders, improve health and prevent diseases. From its -hexane extract, eight compounds were isolated and characterised as: mangiferonic acid (); 1-hydroxymangiferonic acid (), new natural product; mangiferolic acid(); 27-hydroxymangiferolic acid (), reported here for the first time as propolis constituent; 27-hydroxymangiferonic acid (); -amyrin (); -amyrin () and lupeol (). The chemical structures of the isolated compounds were elucidated using spectroscopic methods, such as 1D and 2D-NMR, mass spectrometry and comparison with previous published reports.
