A novel nanobody (Nb)-based voltammetric immunosensor coupled with horseradish peroxidase concatemer-modified hybridization chain reaction (HRP-HCR) signal amplifying system is described to realize the rapid and ultrasensitive detection of AFB. To design such an immunoassay, anti-AFB Nbs with smaller molecular size were coated densely onto the surface of Au nanoparticle-tungsten disulfide-multi-walled carbon nanotubes (AuNPs/WS/MWCNTs) functional nanocomposites as an effective molecular recognition element, whereas AFB-streptavidin (AFB-SA) conjugates were ingeniously bound with biotinylated HCR dsDNA nanostructures as the competitor, amplifier, and signal report element. In the presence of AFB targets, a competitive immunoreaction was performed between the analyte and AFB-SA-labeled HCR (AFB-HCR) platform. Upon the addition of SA-modified polyHRP (SA-polyHRP), AFB-HCR nanostructures containing abundant biotins were allowed to cross-link to a quantity of HRP by streptavidin-biotin chemistry for signal amplification and signal conversion. Under optimal conditions, the immunosensor displayed a good linear correlation toward AFB ranging from 0.5 to 10 ng mL with a sensitivity of 2.7 μA • (mL ng) and an ultralow limit of detection (LOD) of 68 fg mL. The specificity test showed that the AFB immunosensor had no obvious cross-reaction with OTA, DON, ZEN, and FB. The signal of this sensor decreased by 10.18% in 4 weeks indicating satisfactory stability, and its intra- and inter-laboratory reproducibility was 3.42~10.35% and 4.03%~12.11%, respectively. This biosensing system will open up new opportunities for the detection of AFB in food safety and environmental analysis and extend a wide range of applications in the analysis of other small molecules. The development of a sensory signal amplification approach is very crucial for rapid and precise detection of aflatoxin B (AFB). However, such approaches remain scarce due to the weak interactions between AFB and the sensing probes. Herein, the first example of a dual-excitation fluorescent platform for antibody-free AFB detection is reported, which is assembled by an ordered π-π stack of cationic perylene derivative (PTHA) and tris(2,2'-bipyridine)ruthenium(II) [Ru(bpy)].

Background: The study objective was to develop and validate a clinical decision support system (CDSS) to guide clinicians through the diagnostic evaluation of hospitalized individuals with suspected pulmonary tuberculosis (TB) in low-prevalence settings.
Methods: The "TBorNotTB" CDSS was developed using a modified Delphi method. The CDSS assigns points based on epidemiologic risk factors, TB history, symptoms, chest imaging, and sputum/bronchoscopy results.

This study assessed the impact of race and ethnicity on longitudinal test variability and time to detect glaucoma progression using standard automated perimetry (SAP) and optical coherence tomography (OCT). The sample consisted of 47,003 SAP tests from 5402 eyes and 25,480 OCT tests from 4125 eyes, with 20% of participants self-identifying as Black or African American and 80% as White; 29% as Hispanic or Latino and 71% as Not Hispanic or Latino. Variability was measured using standard deviations of residuals from linear regression models for SAP mean deviation (MD) and OCT retinal nerve fiber layer (RNFL) thickness over time.

This work implements a mid-level data fusion methodology on spectral data from handheld X-ray fluorescence and laser-induced breakdown spectroscopy analyzers to quantify plutonium surrogate (CeO) contamination in soil samples for the first time. Spectral data from each analyzer were used independently to train supervised machine learning regressions to predict Ce concentration. Fused features from both data sets were then used to train the same models, comparing prediction performance by evaluating model precision and sensitivity.

A manganese porphyrin wrapped DNA dendrimer (Mn-DD) was developed through enzyme-free DNA self-assembly and simple and mild groove binding of porphyrin. The Mn-DD not only possessed plenty of manganese porphyrin to amplify the chemiluminescence (CL) signal, but also can be modified with diverse groups via DNA hybridization. Combined with an immunosensor array, Mn-DD can be utilized for CL immunoassay of multiple mycotoxins as a universal tag.
