Objectives: We aimed to evaluate the proportional values of maximum standardized uptake value (SUV) for cervical lymph nodes on F-fluorodeoxyglucose (FDG) positron emission tomography/computed tomography (PET/CT) for prediction of the presence of metastasis in patients with larynx squamous cell cancer (LSCC).
Methods: This retrospective study involved 43 patients with LSCC. All patients underwent resection of the primary tumor and neck dissection within 4 weeks after undergoing F-FDG PET/CT examinations. Receiver operating characteristic (ROC) analysis was performed to evaluate the lymph node SUV/primary tumor SUV (SUVLN/SUVPT), lymph node SUV/aortic SUV (SUVLN/SUVA), and lymph node SUV/ liver SUV (SUVLN/SUVL) ratios for diagnosis of lymph node metastasis.
Results: SUVLN/SUVA, SUVLN/SUVL, and SUVLN/SUVPT rates were significantly higher in metastatic lymph nodes compared to non-metastatic nodes. ROC analysis for metastasis showed that the cut-off thresholds were 3.87 for SUVLN; 1.78 for SUVLN /SUVA; 1.08 for SUVLN/SUVL; and 0.36 for SUVLN/SUVPT. The diagnostic sensitivity, specificity and AUC were 83.7%, 77%, 0.856 for SUVLN; 79.7%, 84%, 1.78 for SUVLN/SUVA; 84.1%, 76%, 0.833 for SUVLN/SUVL; and 53.6%, 76%, 0.666 for SUVLN/SUVPT, respectively.
Conclusion: SUVLN/SUVA, SUVLN/SUVL, and SUVLN/SUVPT ratios can be safely used for diagnosis of cervical lymph node metastasis in patients with LSCC.

Despite their safety and widespread use, conventional protein antigen-based subunit vaccines face significant challenges such as low immunogenicity, insufficient long-term immunity, poor CD8 T-cell activation, and poor adaptation to viral variants. To address these issues, an infection-mimicking gel (IM-Gel) is developed that is designed to emulate the spatiotemporal dynamics of immune stimulation in acute viral infections through in situ supramolecular self-assembly of nanoparticulate-TLR7/8a (NP-TLR7/8a) and an antigen with tannic acid (TA). Through collagen-binding properties of TA, the IM-Gel enables sustained delivery and enhanced retention of NP-TLR7/8a and protein antigen in the lymph node subcapsular sinus of mice for over 7 days, prolonging the exposure of vaccine components in both B cell and T cell zones, leading to robust humoral and cellular responses.
