Erlotinib sensitivity of p.D321N mutation in head and neck squamous cell carcinoma. | LitMetric

AI Article Synopsis

  • Head and neck squamous cell carcinoma (HNSCC) currently lacks reliable biomarkers to predict how well patients will respond to drugs.
  • Researchers identified two mutations, p.D321N and p.R135K, in patients with recurrent HNSCC through targeted sequencing, and also noted a previously exceptional responder with mutation p.E322K.
  • The study used computer modeling and drug tests to analyze these mutations, finding that p.D321N significantly increases sensitivity to the drug erlotinib, while the effect of p.R135K is less pronounced.

Article Abstract

Head and neck squamous cell carcinoma (HNSCC) lacks predictive biomarkers for drug responses. By targeted sequencing, we identified two mutations in recurrent HNSCC, p.D321N, and p.R135K. We previously reported an exceptional erlotinib responder with p.E322K. Here, by in silico and drug studies, we determined functions of these two recurrence-associated mutations. Residues D321, R135, and E322 are in 3D proximity. p.D321N drives marked in vivo erlotinib sensitivity, while p.R135K's effect is moderate.
