Apoptosis signal-regulating kinase 1 (ASK1) is a key mediator in the apoptotic and inflammatory cellular stress response. To investigate the therapeutic value of modulating this pathway in neurological disease, we have completed medicinal chemistry studies to identify novel CNS-penetrant ASK1 inhibitors starting from peripherally restricted compounds reported in the literature. This effort led to the discovery of , a novel ASK1 inhibitor with good potency (cell IC = 138 nM), low clearance (rat Cl/Cl = 0.36/6.7 L h kg) and good CNS penetration (rat = 0.38).

This study aims to investigate the effect and potential mechanism of Polygonati Rhizoma aqueous extract in protecting mice from gastric mucosal injury(stomach Yin deficiency). ICR mice were administrated with the mixture of pungent substance extract and alcohol by gavage once a day for 6 weeks to establish the mouse model of gastric mucosal injury with gastric Yin deficiency. The modeled mice were randomized into three groups of model and Polygonati Rhizoma aqueous extract administrated at 0.

GS-4997 halts the progression of tubulointerstitial injury in lupus nephritis.

FASEB J

Department of Nephrology, Hunan Clinical Research Center for Chronic Kidney Disease, Hunan Provincial People's Hospital, The First Affiliated Hospital of Hunan Normal University, Changsha, Hunan, People's Republic of China.

Tubulointerstitial injury has been increasingly recognized as an important component in lupus nephritis (LN) pathology over the last decades. However, current clinical treatment options for this process remain limited. In this study, we aimed to investigate the potential benefits of GS-4997, a selective inhibitor of ASK1, in tubulointerstitial injury of LN.

Fish oocyte maturation (FOM) is a critical biological process that occurs before ovulation and is influenced by gonadotropins, particularly luteinizing hormone (LH). The release of LH stimulates the ovarian follicle to produce a maturation-inducing hormone (MIH), specifically 17α, 20β-dihydroxy-4-pregnen-3-one (17α, 20β-DP), which initiates the formation of maturation-promoting factor (MPF) through the activation of cyclin B and cdc2 kinase. Insulin-like growth factor I (IGF-I) significantly regulates ovarian functions, including steroidogenesis, by activating its membrane receptors and the tyrosine kinase pathway.

Background: Aging of the brain is a major contributor to the onset and progression of neurodegenerative diseases. Conventional treatments for these diseases are often limited by significant side effects and a lack of efficacy in halting disease progression. Ginsenoside compound K (CK), a bioactive secondary metabolite derived from ginseng, has shown promise because of its potent antioxidant properties.
