This manuscript details the modifications made to the HPLC assay method described in the USP monograph for Acetazolamide Compounded Oral Suspension.•The method was modified to allow the quantification of acetazolamide in two new suspension vehicles: Oral Mix and Oral Mix SF;•It was validated for linearity, accuracy, precision and specificity;•It was demonstrated stability-indicating and suitable for use in a stability study using these vehicles.

Glaucoma, a leading cause of irreversible blindness, affects about 70 million people globally. Its treatment focuses on reducing intraocular pressure. Acetazolamide, a potent anti-glaucoma drug, is currently used only systemically due to low solubility and permeation, which cause severe side effects.

Article Synopsis
  • The study aimed to improve the estimation of arterial blood radioactivity concentration (Ca10) for measuring regional cerebral blood flow (rCBF) using machine learning techniques, specifically artificial neural networks (ANN), to avoid invasive blood sampling.
  • It involved analyzing data from 294 patients, with 235 used for training a model to predict Ca10 based on various patient and procedural parameters, and 59 for testing the model's accuracy against traditional methods.
  • Results showed that the proposed ML model yielded higher correlation values (0.81) for Ca10 estimation compared to the conventional method (0.66), showcasing its potential for more reliable assessments of rCBF and cerebral vascular reactivity (CVR).

Carbonic anhydrase inhibitors (CAIs) are prescription drugs also used in doping to dilute urine samples and tamper with urinalyses. Dorzolamide, brinzolamide, and acetazolamide are prohibited by the World Anti-Doping Agency. Detecting CAIs and their metabolites in biological samples is crucial to documenting misuse in doping.

Acetazolamide smartphone-based detection via its competition with sulfamethoxazole on molecularly imprinted polymer: A proof-of-concept.

J Pharm Biomed Anal

September 2022

Laboratoire Génie des Procedés & Environnement, Faculté des Sciences et Techniques, Hassan II University of Casablanca, B.P. 146, Mohammedia, Morocco. Electronic address:

In this work, a competitive assay based on molecularly imprinted polymer (MIP) is developed for the rapid screening of acetazolamide (ACZ) using a colorimetric method coupled with smartphone-based detection. Because of it is an analog of ACZ and its ability to be transformed into a colored product, sulfamethoxazole (SMX) was used as a competitor to indirectly determine ACZ from the inhibition degree of SMX onto the MIP. It was found firstly that ACZ-SMX competition was not possible without using support.
