AI Article Synopsis

  • - Three new inositol angelates and two new tirucallane-type alkaloids were discovered, with their structures identified using advanced techniques like 1D and 2D NMR and HR-ESI-MS.
  • - The compounds showed notable cytotoxic effects against the HepG2 human cancer cell line, with IC values of 8.4 and 13.2 μM, indicating their potential as cancer treatments.
  • - Additionally, the compounds demonstrated significant growth inhibition against larvae, suggesting they may have applications in pest control as well.

Article Abstract

Three new inositol angelate compounds () and two new tirucallane-type alkaloids ( and ) were isolated from the , and their structures were established mainly by means of combination of 1D and 2D nuclear magnetic resonance and HR-ESI-MS. Based on cytotoxicity testing, compounds and exhibited significant cytotoxic activity against human cancer cell line HepG2 with IC value at 8.4 and 13.2 μM. In addition, compounds and also showed remarkable growth inhibitory activity to larvae.

Unlabelled: Visceral leishmaniasis (VL), caused by , remains challenging to treat due to severe side effects and increasing drug resistance associated with current chemotherapies. Our study investigates the anti-leishmanial potential of from Uttarakhand, India, with extracts prepared from leaves and stems using ethanol and hexane. Advanced GC-MS analysis identified over 100 bioactive compounds, which were screened using molecular docking to assess their binding to LdHEL-67, a DDX3-DEAD box RNA helicase of donovani.

First-line immune checkpoint inhibitor (ICI) combinations show responses in subsets of hepatocellular carcinoma (HCC) patients. Nearly half of HCCs are Wnt-active with mutations in (encoding for β-catenin), , or , and demonstrate limited benefit to ICI due to an immune excluded tumor microenvironment. We show significant tumor responses in multiple β-catenin-mutated immunocompetent HCC models to a novel siRNA encapsulated in lipid nanoparticle targeting (LNP-CTNNB1).

Adoptive cell therapy (ACT) can address an unmet clinical need for patients with relapsed/refractory acute myeloid leukemia (AML), but its effect is often modest in the setting of high tumor burden. In this study, we postulated that strategies to lower the AML apoptotic threshold will augment T cell killing of AML cells. BH3 mimetics, such as venetoclax, are a clinically approved class of compounds that predispose cells to intrinsic apoptosis by inhibiting anti-apoptotic mitochondrial proteins.
