Rapid evaporative-ionization mass spectrometry (REIMS) coupled with an electroknife as a sampling device was recently employed in many application fields to obtain a rapid characterization of different samples without any need for extraction or cleanup procedures. In the present research, REIMS was used to obtain a metabolic profiling of the fruit, thus extending the applicability of such a technique to the investigation of phytochemical constituents. In particular, the advantages of REIMS linked to a typical electrosurgical handpiece were applied for a comprehensive screening of this botanical species, by exploiting the mass accuracy and tandem MS capabilities of a quadrupole-time of flight analyzer. Then, 78 biomolecules were positively identified, including phenols, fatty acids and phospholipids. In the last decade, (Lam.) Benth. fruit has attracted special interest for its drug-like properties, e.g., its use for infertility treatments and as anti-tumor agent, as well as against fungal and bacterial infections, diabetes, and inflammatory processes. Many of these properties are currently correlated to the presence of phenolic compounds, also detected in the present study, while the native lipid composition is here reported for the first time and could open new directions in the evaluation of therapeutic activity.

Steak samples were collected from the longissimus lumborum muscles of beef carcasses (Canada AA, n = 1505; Canada AAA, n = 1363) over a 3-year period. Steaks were aged for 14 d, and tenderness was determined by slice shear force (SSF). Metabolomic profiling of beef samples was performed using rapid evaporative ionization mass spectrometry (REIMS) (N = 2853).

Article Synopsis
  • The study utilized Rapid Evaporative Ionization Mass Spectrometry (REIMS) along with chemometric analysis to differentiate between benign and malignant canine mammary tumors (CMTs), achieving high accuracy in tumor identification.
  • Results indicated a significant increase in phospholipid synthesis in malignant tumors compared to normal mammary tissues, highlighting specific phospholipid species as key indicators of carcinogenesis.
  • The research also uncovered a link between cancer and inflammation through the dysregulation of fatty acid metabolism, suggesting that understanding these metabolic changes could enhance diagnostic and treatment strategies for tumors.

Some cancers such as glioblastoma (GBM), show minimal response to medical interventions, often only capable of mitigating tumor growth or alleviating symptoms. High metabolic activity in the tumor microenvironment marked by immune responses and hypoxia, is a crucial factor driving tumor progression. The many developments in mass spectrometry (MS) over the last decades have provided a pivotal tool for studying proteins, along with their posttranslational modifications.

Identification strategy of wild and cultivated Astragali Radix based on REIMS combined with two-dimensional LC-MS.

NPJ Sci Food

November 2024

State Key Laboratory of Natural and Biomimetic Drugs, School of Pharmaceutical Sciences, Peking University, Beijing, China.

A rapid and real-time method was established based on the combination of rapid evaporative ionization mass spectrometry (REIMS) and two-dimensional liquid chromatography mass spectrometry (2DLC-MS) for identification of wild Astragali Radix (WAR) and cultivated AR (CAR). The samples were analyzed under ambient ionization without time-consuming sample preparation. The phenotypic data of WAR and CAR were used to develop a real-time recognition model.

Building extensive drug candidate libraries as early in the development pipeline as possible, with high-throughput in vitro absorption, distribution, metabolism, and excretion (ADME) profiling, is crucial for the selection of lead compounds to guide subsequent research and production phases. Traditionally, the analysis of metabolic stability assays heavily relies on high-throughput LC-MS/MS (liquid chromatography tandem mass spectrometry) techniques to meet with the lead profiling demands. Laser-assisted rapid evaporative ionization mass spectrometry (LA-REIMS) is a quick and efficient technique for characterizing complex biological samples without laborious sample preparation.
