The Type III Secretion Systems (T3SS) needle complex is a conserved syringe-shaped protein translocation nanomachine with a mass of about 3.5 MDa essential for the survival and virulence of many Gram-negative bacterial pathogens. This system is composed of a membrane-embedded basal body and an extracellular needle that deliver effector proteins into host cells. High-resolution structures of the T3SS from different organisms and infection stages are needed to understand the underlying molecular mechanisms of effector translocation. Here, we present the cryo-electron microscopy structure of the isolated Shigella T3SS needle complex. The inner membrane (IM) region of the basal body adopts 24-fold rotational symmetry and forms a channel system that connects the bacterial periplasm with the export apparatus cage. The secretin oligomer adopts a heterogeneous architecture with 16- and 15-fold cyclic symmetry in the periplasmic N-terminal connector and C-terminal outer membrane ring, respectively. Two out of three IM subunits bind the secretin connector via a β-sheet augmentation. The cryo-EM map also reveals the helical architecture of the export apparatus core, the inner rod, the needle and their intervening interfaces.

Understanding human anatomy is crucial for improving public health outcomes; however, effective methods of engaging the public in this domain remain underexplored. This report investigates four hands-on, creative, and accessible methods for enhancing anatomical knowledge during public engagement events: drawing, clay modeling, needle felting, and baking. Drawing on the principles of the Portal to Public Framework and adult learning theory, we explore how each method offers ethical and inexpensive opportunities for interactive learning, devoid of complex health, and safety and ethical concerns.

This study used oxygen sensors coupled with amperometry to examine changes in brain (nucleus accumbens) and peripheral (subcutaneous space) oxygen levels following intravenous (IV) and intraperitoneal (IP) fentanyl injections at different doses. To avoid stress associated with traditional IP injection, fentanyl was delivered via chronically implanted IP catheter. SC oxygen levels decreased in a dose-dependent monophasic manner, whereas brain oxygen responses were more complex, showing modest increases at low fentanyl doses (<3 μg/kg for IV and <10 μg/kg for IP) and biphasic effects (decrease followed by increase) at higher doses (≥10 μg/kg for IV and ≥270 μg/kg for IP).

Transseptal puncture (TSP) is widely used in catheter-based cardiac interventions to gain left atrial (LA) access, but its workflow has remained largely unchanged and is still a source of serious complications. Pulsed field ablation (PFA) for pulmonary vein isolation (PVI) has been shown to be at least comparable with radiofrequency ablation (RFA) in terms of safety and efficacy. However, PFA catheter delivery to the LA typically requires a standard TSP and an over the wire sheath exchange which can limit workflow and lengthen procedure time-a shorter procedure time being a proposed advantage of PFA over RFA.

Traditional mRNA vaccine formulation loaded by lipid nanoparticle (mRNA-LNP) has several shortcomings in clinical application, including the need for cryopreservation, discomfort associated with intramuscular injections, and the risk of liver aggregation. Dissolvable microneedles (DMNs), as a novel transdermal drug delivery platform, can overcome the skin barrier to deliver drugs directly into the skin in a minimally invasive manner. However, mRNA-LNP is unstable and easily degraded during the solidification of DMN.
