The presence of cell surface protein CD47 allows cancer cells to evade innate and adaptive immune surveillance resulting in metastatic spread. CD47 binds to and activates SIRPα on the surface of myeloid cells, inhibiting their phagocytic activity. On the other hand, CD47 binds the matricellular protein Thrombospondin-1, limiting T-cell activation.

The CDK4/6 Inhibitor Palbociclib Inhibits Estrogen-Positive and Triple Negative Breast Cancer Bone Metastasis In Vivo.

Cancers (Basel)

April 2023

CDK 4/6 inhibitors have demonstrated significant improved survival for patients with estrogen receptor (ER) positive breast cancer (BC). However, the ability of these promising agents to inhibit bone metastasis from either ER+ve or triple negative BC (TNBC) remains to be established. We therefore investigated the effects of the CDK 4/6 inhibitor, palbociclib, using in vivo models of breast cancer bone metastasis.

Genomic screening reveals ubiquitin-like modifier activating enzyme 1 as a potent and druggable target in c-MYC-high triple negative breast cancer models.

Article Synopsis
  • Triple negative breast cancer (TNBC) is responsible for over 30% of breast cancer deaths, yet it represents only 10-15% of all cases, highlighting a significant challenge in treatment.
  • Research using CRISPR/Cas9 screening identified that TNBC cells are particularly sensitive to the depletion of UBA1, a ubiquitin-like modifier activating enzyme.
  • The UBA1 inhibitor TAK-243 induces severe ER stress and cell death in TNBC cells, particularly those with high c-MYC levels, showing promise for shrinking tumors and reducing metastasis in both lab models and patient-derived xenografts.

Behcet's disease is a multi-systemic inflammatory disease with a clinical spectrum as a triple complex of recurrent oral, genital ulcers, and uveitis. Cardiac involvement in patients with Behcet's disease is extremely rare and often associated with poor prognosis. Behcet's disease should be considered in the differential diagnosis of right ventricular mass especially in young adults, even there is no typical clinical features of Behcet's disease.
