Experience-Dependent Development and Maintenance of Binocular Neurons in the Mouse Visual Cortex.

The development of neuronal circuits requires both hard-wired gene expression and experience-dependent plasticity. Sensory processing, such as binocular vision, is especially sensitive to perturbations of experience. We investigated the experience-dependent development of the binocular visual cortex at single-cell resolution by using two-photon calcium imaging in awake mice. At eye-opening, the majority of visually responsive neurons are monocular. Binocular neurons emerge later with visual experience and acquire distinct visual response properties. Surprisingly, rather than mirroring the effects of visual deprivation, mice that lack the plasticity gene Arc show increased numbers of binocular neurons and a shift in ocular dominance during development. Strikingly, acutely removing Arc in the adult binocular visual cortex also increases the number of binocular neurons, suggesting that the maintenance of binocular circuits requires ongoing plasticity. Thus, experience-dependent plasticity is critical for the development and maintenance of circuits required to process binocular vision.

However, new research in fruit fly (Drosophila) binocular vision reveals ultrafast photomechanical photoreceptor movements significantly enhance information processing, thereby impacting a fly's perception of its environment and behaviour. The coding advantages resulting from these mechanical processes suggest that similar physical motion-based coding strategies may affect neural communication ubiquitously.

Homeobox transcription factor Engrailed1 (En1) is expressed in the ectoderm and mediates the establishment of retinotectal topography, but its role in eye-specific retinogeniculate segregation and visual function remains unclear. Parvalbumin (PV) neurons, which are widely distributed in the visual pathway, play a crucial role in visual development and function. In this study, we conditionally knocked out En1 gene in PV neurons and found an expansion of the ipsilateral eye projection, while no significant effects were observed in the contralateral eye projection.

Due to technical roadblocks, it is unclear how visual circuits represent multiple features or how behaviorally relevant representations are selected for long-term memory. Here we developed Moculus, a head-mounted virtual reality platform for mice that covers the entire visual field, and allows binocular depth perception and full visual immersion. This controllable environment, with three-dimensional (3D) corridors and 3D objects, in combination with 3D acousto-optical imaging, affords rapid visual learning and the uncovering of circuit substrates in one measurement session.
