Background: Annexin A3 (Anxa3) is a member of the calcium-regulated, cell membrane-binding family of annexin proteins. We previously confirmed that Anxa3 is expressed in the endothelial lineage in vertebrates and that loss of anxa3 in Xenopus laevis leads to embryonic blood vessel defects. However, the biological function of Anxa3 in mammals is completely unknown. In order to investigate Anxa3 vascular function in mammals, we generated an endothelial cell-specific Anxa3 conditional knockout mouse model (Anxa3 ;Tie2-Cre).

Results: Anxa3 ;Tie2-Cre mice are born at Mendelian ratios and display morphologically normal blood vessels during development. However, loss of Anxa3 leads to artery-vein (AV) misalignment characterized by atypical AV crossovers in the postnatal and adult retina.

Conclusions: Anxa3 is not essential for embryonic blood vessel formation but is required for proper parallel AV alignment in the murine retina. AV crossovers associated with Anxa3 ;Tie2-Cre mice are similar to AV intersections observed in patients with branch retinal vein occlusion (BRVO), although we did not observe occluded vessels. This new Anxa3 mouse model may provide a basis for understanding AV crossover formation associated with BRVO.

State Key Laboratory of Ophthalmology, Optometry and Visual Science, Eye Hospital, Wenzhou Medical University, Wenzhou, China.

Purpose: Protein arginine methyltransferase 1 (PRMT1) is an integral constituent of numerous cellular processes. However, its role in corneal epithelial wound healing (CEWH) remains unclear. This study investigates the impact of PRMT1 on cellular mechanisms underlying corneal epithelial repair and its potential to improve wound healing outcomes.

Objective: To identify neutrophil extracellular traps (NETs)-related molecular clusters and establish a novel gene signature for predicting biochemical recurrence in prostate cancer (PCa).

Methods: The transcriptome and clinicaldata of PCa sampleswere obtained from The TCGA and GEO databases. To identify NET-related molecular clusters, consensus clustering analyses were performed.

Identification of Spatial Specific Lipid Metabolic Signatures in Long-Standing Diabetic Kidney Disease.

November 2024

Key Laboratory of Phytochemistry and Natural Medicines, Dalian Institute of Chemical Physics, Chinese Academy of Sciences, Dalian 116023, China.

Diabetic kidney disease (DKD) is a major complication of diabetes leading to kidney failure. This study investigates lipid metabolism profiles of long-standing DKD (LDKD, diabetes duration > 10 years) by integrative analysis of available single-cell RNA sequencing and spatial multi-omics data (focusing on spatial continuity samples) from the Kidney Precision Medicine Project. Two injured cell types, an injured thick ascending limb (iTAL) and an injured proximal tubule (iPT), were identified and significantly elevated in LDKD samples.

Article Synopsis
  • Sepsis is a serious condition caused by an uncontrolled immune response to infection, leading to organ failure and high death rates, but current diagnostic methods for it are not very effective.
  • This study explores new potential biomarkers for sepsis by analyzing multiple datasets to identify genes that could help improve diagnosis, culminating in the identification of two specific genes.
  • One of these genes showed promise as a diagnostic tool, accurately distinguishing between different types of sepsis and exhibiting notable changes in expression levels in a mouse model of sepsis.
