17β-Estradiol (E2) action can be mediated by the full-length estrogen receptor alpha (ERα66), and also by the AF1 domain-deficient ERα (ERα46) isoform, but their respective sensitivity to E2 is essentially unknown. We first performed a dose response study using subcutaneous home-made pellets mimicking either metestrus, proestrus or a pharmacological doses of E2, which resulted in plasma concentrations around 3, 30 and 600 pM, respectively. Analysis of the uterus, vagina and bone after chronic exposure to E2 demonstrated dose-dependent effects, with a maximal response reached at the proestrus-dose in wild type mice expressing mainly ERα66. In contrast, in transgenic mice harbouring only an ERα deleted in AF1, these effects of E2 were either strongly shifted rightward (10-100-fold) and/or attenuated, depending on the tissue studied. Finally, experiments in different cell lines transfected with ERα66 or ERα46 also delineated varying profiles of ERα AF1 sensitivity to E2. Altogether, this work emphasizes the importance of dose in the tissue-specific actions of E2 and demonstrates the key sensitizing role of AF1 in ERα activity.

Although therapies based on direct-acting antivirals (DAAs) effectively eradicate hepatitis C virus (HCV) in patients, there is still a high risk of liver fibrosis even after a sustained virological response. Therefore, it is of great clinical importance to understand the mechanism of potential factors that promote liver fibrosis after virological cure by treatment with DAAs. Here, we found that tubulointerstitial nephritis antigen-like 1 (TINAGL1) is significantly increased in HCV-infected hepatocytes and in the liver of patients with liver fibrosis, and that higher TINAGL1 expression persists in HCV-eradicated hepatocytes after treatment with DAAs.

Herein, we discuss the idea that fluorescent materials/molecules should logically show potential photoelectrochemistry (PEC) activity, and, in particular, the PEC of fluorescent small molecules (previously usually acting only as dye sensitizers for conventional semiconductors) is explored. After examining the PEC activities of some typical inorganic or organic fluorescent materials/molecules and by adopting methyl violet (MV) with the highest PEC activity among the examined fluorescent small molecules, a new and efficient (MV/Au nanoparticles (AuNPs))/fluorine-doped tin oxide (FTO) photoanode without conventional semiconductor(s) is prepared by layer-by-layer alternating the electrodeposition of AuNPs and the adsorption of MV. A bilirubin oxidase (BOD)/CuCoO/FTO bio-photocathode is prepared by electrodeposition, calcination and cast-coating.

The development of narrowband emissive, bright, and stable solution-processed organic light-emitting diodes (SP-OLEDs) remains a challenge. Here, a strategy is presented that merges within a single emitter a TADF sensitizer responsible for exciton harvesting and an MR-TADF motif that provides bright and narrowband emission. This emitter design also shows strong resistance to aggregate formation and aggregation-cause quenching.
