Calreticulin arms NK cells against leukemia.

Published: July 2021

Calreticulin (CALR) exposed on the surface of cancer cells succumbing to therapy delivers robust phagocytic signals that support the activation of adaptive anticancer immune responses. Recent data from our group demonstrate that spontaneous CARL exposure on leukemic blasts also supports innate anticancer immunity by natural killer (NK) cells via an indirect mechanism relying on myeloid CD11cCD14 cells.

Background: Root hairs are single-celled projections on root surfaces, critical for water and nutrient uptake. Here, we describe the first short root hair mutant in wheat ( L.), identified in a mutagenized population and termed here short root hair 1 ().

A genome-wide CRISPR/Cas9 screen identifies calreticulin as a selective repressor of ATF6α.

Activating transcription factor 6 (ATF6) is one of three endoplasmic reticulum (ER) transmembrane stress sensors that mediate the unfolded protein response (UPR). Despite its crucial role in long-term ER stress adaptation, regulation of ATF6 alpha (α) signalling remains poorly understood, possibly because its activation involves ER-to-Golgi and nuclear trafficking. Here, we generated an ATF6α/Inositol-requiring kinase 1 (IRE1) dual UPR reporter CHO-K1 cell line and performed an unbiased genome-wide CRISPR/Cas9 mutagenesis screen to systematically profile genetic factors that specifically contribute to ATF6α signalling in the presence and absence of ER stress.

Article Synopsis
  • A study assessed the outcomes of patients with non-muscle invasive bladder cancer (NMIBC) who had transurethral resection using either sterile water or glycine for irrigation, focusing on tumor recurrence and progression.
  • Researchers reviewed records of 530 patients and found that after matching, those treated with sterile water had significantly lower rates of recurrence (43%) and progression (85%) compared to those treated with glycine (71% and 94%, respectively).
  • The study concluded that sterile water is linked to higher recurrence and progression rates, likely due to a weaker immune response compared to glycine, which induced more markers of immunogenic cell death.

Optical biosensor based on weak measurement for ultra-sensitive detection of calreticulin in human serum.

A novel real-time optical phase sensing method based on the Mach-Zehnder interference principle has been proposed for the detection of calreticulin (CRT) levels in human serum samples. In this approach, anti-CRT antibodies are utilized to capture CRT molecules in serum, leading to a phase shift in both the measuring and reference arms of the system. By employing the concept of weak amplification within the framework of weak measurements, it becomes feasible to continuously monitor the response of CRT in real-time, allowing for the precise determination of serum CRT content at the picomolar level.
