Article Synopsis
  • Previous research demonstrated that brucine dissolving-microneedles (Bru-DMNs) effectively treat rheumatoid arthritis (RA) while addressing issues common with oral and systemic drug delivery.
  • This study developed a novel method for continuous sampling and analysis of drug distribution in RA rats, revealing that Bru-DMNs achieve better retention in joint cavities than in blood.
  • The findings highlight a complex PK-PD relationship for Bru against RA, offering insights into how transdermal drugs may work and their potential side effects.

Toxic alkaloids are typically found in herbal medicines and have strong pharmacological effects and a broad therapeutic spectrum. On the other hand, toxic alkaloids exert toxicological activities in vivo; as such they have a narrow therapeutic window and can induce poisoning due to incorrect dose or misuse. In this view, there is an urgent need to develop a rapid and sensitive assay to detect these toxic alkaloids.

Glycine Receptors (GlyRs) provide inhibitory neuronal input in the spinal cord and brainstem, which is critical for muscle coordination and sensory perception. Synaptic GlyRs are a heteromeric assembly of α and β subunits. Here we present cryo-EM structures of full-length zebrafish α1βGlyR in the presence of an antagonist (strychnine), agonist (glycine), or agonist with a positive allosteric modulator (glycine/ivermectin).
