Fibrin elasticity and coagulation.

Sudden increase of viscosity in former times indicated the start of coagulation. Yet its measurement destroyed the structure of coagulum. By the precursor method of thrombelastography the author 1944 found elasticity to be the essential physiological property of coagulum. In the production of elastic fibrin structure its early phase is the most efficient in this respect. The speed of prime structure formation is extremely fast in presence of enough phospholipid as well as of plasma factor XIII. Even high amounts of thrombin cannot replace one or both of these substances indispensable to grow rapidly a perfect fibrin web. This phase yet does not become effective if it is not accompanied by the orbital micro-flow of the new orbitometry method. Its shear is comparable to that in a coronary artery. The special resonance effect of the method in combination with the early phase of fibrin production is generating some kind of a physiological feed back: increasing fibrin will strengthen shear stress as long as less platelets are entangled which will reduce the elastic flexibility of fibrin web. Some kind of a "coagulation spin effect" in optimal combination of shear stress, phospholipid and factorXIII, will extremely fast originate a firm fibrin structure, a mechanism which may be of significance for a fast occlusion of arterial stenoses.

Atrial fibrillation (AF) and atrial myopathy are recognized contributors to cardiovascular morbidity, particularly ischemic stroke. AF poses an elevated risk of thrombogenesis due to irregular heart rhythm leading to blood stasis and clot formation. In Vitro Model of Vascular Remodeling Under Microfluidic Perfusion.

We developed a portable microfluidic system that combines spontaneous lumen formation from human umbilical endothelial cells (HUVECs) in fibrin-collagen hydrogels with active perfusion controlled by a braille actuator. Adaptive interstitial flow and feedthrough perfusion switching enabled the successful culture of spontaneously formed naturally branched lumens for more than one month. We obtained many large-area (2 mm × 3 mm) long-term (more than 30 days per run) time-lapse image datasets of the in vitro luminal network using this microfluidic system.

Large skeletal muscle injuries such as volumetric muscle loss (VML) disrupt native tissue structures, including biophysical and biochemical signaling cues that promote the regeneration of functional skeletal muscle. Various biofabrication strategies have been developed to create engineered skeletal muscle constructs that mimic native matrix and cellular microenvironments to enhance muscle regeneration; however, there remains a need to create scalable engineered tissues that provide mechanical stability as well as structural and spatiotemporal signaling cues to promote cell-mediated regeneration of contractile skeletal muscle. We describe a novel strategy for bioprinting multifunctional myoblast-loaded fibrin microthreads (myothreads) that recapitulate the cellular microniches to drive myogenesis and aligned myotube formation.
