Background: Apoptosis is a normal physiological process in organs development, but excessive apoptosis is pathological and harmful. In previous studies, apoptosis-related genes are considered to be involved in the onset of osteoarthritis, but the mechanism is unclear. Therefore, we selected two common polymorphisms of apoptosis-related genes ( -248G>A, -717C>A) to explore the relationship with osteoarthritis.

Methods: The two polymorphisms were genotyped by polymerse chain reaction-restriction fragment length polymorphism (PCR-RFLP) in 134 cases with osteoarthritis and 142 controls. These genotypes distributions in controls were checked whether conformed to Hardy-Weinberg equilibrium (HWE). χ test was used to calculate odds ratio (OR) with corresponding 95% confidence interval (95% CI) which evaluated the strength of association between gene polymorphism and osteoarthritis susceptibility.

Results: The χ test showed that genotype frequencies of -248G>A (rs4645878), -717C>A (rs2279115) polymorphisms in control group were consistent with HWE. In -248G>A polymorphism, compared with mutant genotype GA+AA, the common genotype GG increased the susceptibility to osteoarthritis significantly (OR=1.84, 95% CI=1.13-3.00), so G allele was (OR=1.76, 95% CI=1.16-2.68). The homozygous mutant genotype AA in -717C>A carriers easily suffered from osteoarthritis in some condition, compared with CC genotype (=0.03). A allele also increased 0.43 times risk of osteoarthritis development than C allele (OR=1.43, 95% CI=1.02-2.00).

Conclusions: -248G>A and -717C>A polymorphisms may be the independent risk factors for the development of osteoarthritis.

Department of Endocrinology and Metabolism, Zhongshan Hospital, Fudan University, 180 Fenglin Road, Shanghai, 200032, People's Republic of China.

Background: To explore the complex interactions between the tumor microenvironment (TME) of pancreatic ductal adenocarcinoma (PDAC) and the loss of β-cell mass, further elucidating the mechanisms of type 3c diabetes mellitus (T3cDM) onset.

Methods: Single-cell RNA sequencing was employed to analyze the PDAC TME, identifying cell interactions and gene expression changes of endocrine cells. Pathological changes and paraneoplastic islets were assessed in the proximal paratumor (PP) and distal paratumor (DP).

Diquat (DQ) is a pro-oxidant that generates free radicals in cells through redox reactions, leading to the induction of oxidative stress. During the processes of growth and reproduction, poultry are particularly vulnerable to oxidative stress. Selenium yeast (SeY) serves as an organic selenium source characterized by high activity and low toxicity, imparting antioxidant effects.

Tangerine peel is a traditional Chinese herb and has been widely applied in foods and medicine for its multiple pharmacological effects. Erythropoietin receptor (EPOR), a member of the cytokine receptor family, is widely expressed in multiple tissues in especial kidney and plays protective effects in adverse physiological and pathological conditions. We hypothesized that it might be EPOR agonists existing in Tangerine peel bring such renal benefits.

Organoids are stem cell-derived three-dimensional tissue cultures composed of multiple cell types that recapitulate the morphology and functions of their in vivo counterparts. Organ-on-a-chip devices are tiny chips with interconnected wells and channels designed using a perfusion system and microfluidics to precisely mimic the in vivo physiology and mechanical forces experienced by cells in the body. These techniques have recently been used to reproduce the structure and function of organs in vitro and are expected to be promising alternatives for animal experiments in the future.

Doxorubicin (DOX) is a widely used antitumor drug; however, its use is limited by the risk of serious cardiotoxicity. Dehydroevodiamine (DHE) is a quinazoline alkaloid which has antiarrhythmic effects. The aim of this study was to investigate the protective effect of DHE on doxorubicin-induced cardiotoxicity (DIC) and its potential mechanism.
