Driving in patients with dementia.

J Am Geriatr Soc

Dementia Research Clinic, Johns Hopkins Hospital, Baltimore, Maryland 21205.

Published: December 1988

These pilot data suggest that driving among individuals with incapacitating dementing illness may be an unrecognized, potentially serious problem. A simple 20-item screening questionnaire was administered to 72 referrals to the Dementia Research (outpatient) Clinic of the Johns Hopkins Hospital. Thirty percent of the patient sample had at least one accident since the onset of symptoms of dementia. An additional 11% were reported by caregivers to have "caused" accidents.

Elevated protein lactylation promotes immunosuppressive microenvironment and therapeutic resistance in pancreatic ductal adenocarcinoma.

J Clin Invest

January 2025

Department of Hepatobiliary and Pancreatic Surgery, The First Affiliated Hospital, School of Medicine, Zhejiang University, Hangzhou, China.

Metabolic reprogramming shapes tumor microenvironment (TME) and may lead to immunotherapy resistance in pancreatic ductal adenocarcinoma (PDAC). Elucidating the impact of pancreatic cancer cell metabolism in the TME is essential to therapeutic interventions. "Immune cold" PDAC is characterized by elevated lactate levels resulting from tumor cell metabolism, abundance of pro-tumor macrophages, and reduced cytotoxic T cell in the TME.

Cancers of the brain and central nervous system: global patterns and trends in incidence.

J Neurooncol

January 2025

Cancer Surveillance Branch, International Agency for Research On Cancer (IARC), 25 Avenue Tony Garnier, CS 90627, 69366 LYON CEDEX 07, Lyon, France.

Background: Global comparisons of the burden and impact of cancers of the brain and central nervous system (CNS) are critical for developing effective control strategies and generating etiological hypotheses to drive future research.

Methods: National incidence estimates were obtained from GLOBOCAN 2022, and recorded incidence data from the Cancer in Five Continents series, both developed and compiled by the International Agency for Research on Cancer. We examined the estimated age-standardized incidence rates in 185 countries, as well as time trends in recorded incidence in 35 countries, quantifying the direction and change in the magnitude of the rates using the estimated average percentage change (EAPC).

Background: Although invasiveness is one of the major determinants of the poor glioblastoma (GBM) outcome, the mechanisms of GBM invasion are only partially understood. Among the intrinsic and environmental processes promoting cell-to-cell interaction processes, eventually driving GBM invasion, we focused on the pro-invasive role played by Extracellular Vesicles (EVs), a heterogeneous group of cell-released membranous structures containing various bioactive cargoes, which can be transferred from donor to recipient cells.

Methods: EVs isolated from patient-derived GBM cell lines and surgical aspirates were assessed for their pro-migratory competence by spheroid migration assays, calcium imaging, and PYK-2/FAK phosphorylation.

Generalizability of Randomized Controlled Trials to Routine Clinical Care in Ulcerative Colitis.

Inflamm Bowel Dis

January 2025

Inflammatory Bowel Disease Unit, Division of Gastroenterology and Hepatology, Department of Medicine, University of Calgary, 5th Floor Cal Wenzel Precision Health Building, 3280 Hospital Drive NW, Calgary, AB T2N 4Z6, Canada.

Background: Historically, randomized controlled trials (RCTs) have been criticized for being poorly generalizable to patients with ulcerative colitis (UC) evaluated in routine care. We aimed to evaluate the proportion of patients with UC starting an advanced therapy who would be eligible to participate in phase 3 registrational UC RCTs.

Methods: We conducted a retrospective cohort analysis of UC patients starting vedolizumab, ustekinumab, or tofacitinib at 2 IBD clinics at the University of Calgary.

ISB 1442 is a bispecific biparatopic antibody in clinical development to treat hematological malignancies. It consists of two adjacent anti-CD38 arms targeting non-overlapping epitopes that preferentially drive binding to tumor cells and a low-affinity anti-CD47 arm to enable avidity-induced blocking of proximal CD47 receptors. We previously reported the pharmacology of ISB 1442, designed to reestablish synthetic immunity in CD38+ hematological malignancies.
