A nitroreductase (NTR) responsive fluorescent probe, Na-NO, comprising p-nitrobenzyl as the unique recognition group and 1,8-naphthalimide as fluorophore, was synthesized. Na-NO showed remarkable fluorescence "turn-on" signal in the presence of NTR under DMSO/HO (1:19, v/v) buffered with PBS (pH = 7) solution in the presence of NADH (300 µM). Furthermore, the probe has a low detection limit down to 3.4 ng/mL and it is very sensitive towards the NTR in Escherichia coli (E. coli), Staphylococcus aureus (S. aureus), normal and tumor cells such as HL-7702, HepG-2 and MCF-7.

A sensitized dual-response ratiometric fluorescent sensor integrated smartphone platform for accurate discrimination and detection of tetracycline (TC) homologues was fabricated based on N-CDs-Eu complex. In the sensing system, N-CDs act as a sensitizer of Eu and significantly enhance the fluorescence of TC-Eu complex approximate 40-fold owing to the synergistic effect of antenna effect (AE) and fluorescence resonance energy transfer (FRET). A paper sensor integrated with a smartphone platform is further fabricated for on-site measurement of TC.

CRISPR/Cas 12a system based nucleic acid and non-nucleic acid targets detection faces two challenges including (1) multiple crRNAs are needed for multiple biomarkers detection and (2) insufficient sensitivity resulted from photobleaching of fluorescent dyes and the low kinetic cleavage rate for a traditional single-strand (ssDNA) reporter. To address these limitations, we developed a programmable DNA nanoswitch (NS)-regulated plasmonic CRISPR/Cas12a-gold nanostars (Au NSTs) reporter platform for detection of nucleic acid and non-nucleic acid biomarkers with the assistance of the spatial confinement effect. Through simply programming the target recognition sequence in NS, only one crRNA is required to detect both nucleic acid and non-nucleic acid biomarkers.
