Computational generation of cyclic peptide inhibitors using machine learning models requires large size training data sets often difficult to generate experimentally. Here we demonstrated that sequential combination of Random Forest Regression with the pseudolikelihood maximization Direct Coupling Analysis method and Monte Carlo simulation can effectively enhance the design pipeline of cyclic peptide inhibitors of a tumor-associated protease even for small experimental data sets. Further studies showed that such -evolved cyclic peptides are more potent than the best peptide inhibitors previously developed to this target.

Introduction Transitioning from a medical student to a foundation doctor presents numerous challenges, particularly in managing on-call duties that require quick decision-making, clinical skills, and prioritisation under pressure. The Simulation On-Call (SOC) program was developed as a one-day, immersive simulation event to equip final-year medical students with the skills and confidence needed for these responsibilities. Surgeons in training have a limited opportunity to practice their skills in a realistic surgical environment. Surgical training models are expensive and are often single-use exercises involving consumable supplies. The goal of the work presented in this paper is to increase surgeon training time by developing a surgical training simulator that is portable, inexpensive, and offers a realistic surgical environment.

Background: Otoscope examinations are a fundamental skill in pediatric care, crucial for diagnosing and managing ear conditions such as otitis media. Traditional training methods for pediatric otoscopic examination often rely on adult standardized patients (SPs) or simulated models, which may not be adequate for pediatric examinations.
Objectives: This study evaluates the feasibility and effectiveness of use of children as SPs in Objective Structured Clinical Examinations (OSCEs) to assess medical students' competency in pediatric otoscopy.
Fatigue cracking of rib-to-deck conventional single-sided welded joints is a prevalent issue in orthotropic steel decks (OSDs), significantly impacting their structural integrity and durability. Rib-to-deck innovative double-sided welded joints have the potential to enhance the fatigue resistance of OSD.
