Objective: To investigate the expression level of silencing information regulatory 2 (SIRT2) in cervical cancer and CIN tissues and its clinical significance.

Methods: Immunohistochemistry was carried out to examine the expression of SIRT2 in 262 cases of cervical cancer tissues, 75 cases of precancerous lesions and 75 cases of normal cervical tissues, and the clinical implications of SIRT2 was further analyzed. The protein expression level of SIRT2 in 40 cervical cancer fresh tissue samples and 20 normal cervical tissue samples was detected by Western blot. shRNA-2 virus was transfected into HeLa cells to obtain the SIRT2-down-regulated HeLa cells. And the influence of down-regulation of SIRT2 on cell proliferation and migration was investigated by MTT and Scratch test.

Results: Immunohistochemical results showed that SIRT2 protein expression level gradually increased in cervical cancer, CIN and normal cervix tissues ( < 0.001). Western blot confirmed that SIRT2 expression level was higher in cervical cancer tissues than in normal cervix tissues ( < 0.001). The expression level of SIRT2 in cervical cancer was correlated with some factors, such as lymph node metastasis, histological type, clinical staging and HPV infection ( < 0.05). At the same time, it was independent of some factors including age, differentiation degree, pelvic metastasis, whether involving the cervical interstitial and the involvement depth and whether involving the neck junction and vaginal ends (>0.05). Mortality in SIRT2 high expression patient group was higher than that of SIRT2 low expression patient group ( < 0.05). shRNA interference technique could effectively inhibit the expression level of SIRT2 in HeLa cells ( < 0.05). MTT assay and Scratch test indicated that down-regulation of SIRT2 expression inhibited the proliferation and migration of HeLa cells ( < 0.05).

Conclusion: SIRT2 may play a promoting role in the progress of cervical cancer, and SIRT2 may be related to the development of malignant degree of cervical cancer, and the inhibition of SIRT2 expression may be a potential therapeutic target for cervical cancer.

This study explores the effects of plant compounds on human papillomavirus (HPV)-induced W12 cervical precancer cells and bioelectric signaling. The aim is to identify effective phytochemicals, both individually and in combination, that can prevent and treat HPV infection and HPV associated cervical cancer. Phytochemicals were tested using growth inhibition, combination, gene expression, RT PCR, and molecular docking assays.

Tea is a significant source of flavonoids in the diet. Due to different production processes, the amount of bioactive compounds in unfermented (green) and (semi-)fermented tea differs. Importantly, green tea has a similar composition of phenolic compounds to fresh, unprocessed tea leaves.

Downregulation of Ezrin Suppresses Migration Potential in Cervical Cancer Cells.

Department of Histology and Embryology, Faculty of Medicine, Collegium Medicum in Bydgoszcz, Nicolaus Copernicus University in Toruń, Karłowicza 24, 85-092 Bydgoszcz, Poland.

Background: The literature reports that ezrin (EZR) is important as a linker between microfilaments and cellular environments. Moreover, it affects cancer cell migration, but the exact mechanism is not fully understood. In this study, we aimed to investigate the role of EZR in the migration of two different types of cervical cancer cells-from primary lesion (SiHa) and lymph node metastases (HT-3).

Surgical intervention in asymptomatic retrosternal goiter (RSG) is debated in the absence of suspicious cytology, while performing fine-needle aspiration (FNA) is challenging in thyroids with mediastinal extension. The rate of unexpected thyroid cancers found at the time of thyroidectomy varies widely, while the notion of increased cancer incidence in RSG with respect to cervical goiters is still controversial. We retrospectively reviewed 411 patients with a preoperative diagnosis of multinodular goiter (MNG) (114 retrosternal, 297 cervical) who underwent thyroidectomy at an academic endocrine surgery referral center between January 2019 and October 2022.

Cervical cancer poses a substantial threat to women's health, underscoring the necessity for effective therapeutic agents with low toxicity that specifically target cancer cells. As cancer progresses, increased glucose consumption causes glucose scarcity in the tumor microenvironment (TME). Consequently, it is imperative to identify pharmacological agents capable of effectively killing cancer cells under conditions of low glucose availability within the TME.
